软骨多糖在制备肿瘤疫苗中的初探
Study of Cartilage Polysaccharide on Preparation of Tumor Vaccine
【摘要】 通过建立动物模型优化疫苗制备条件,检测小鼠的免疫器官和脾淋巴细胞增殖情况,用SDS-聚丙烯酰胺凝胶电泳检测L1210细胞蛋白变化,研究了软骨多糖(PS)在制备肿瘤疫苗中的应用。研究表明,培养48h的软骨多糖和细胞共混合物(PS&cell48h)对健康小鼠的免疫效果最好,使小鼠的免疫器官得到保护,并且显著提高了小鼠的体液免疫,对细胞免疫也有较好的刺激作用,通过细胞免疫荧光检测到特异性的抗体,推测可能是软骨多糖诱导48h后抗原蛋白的变化使小鼠机体产生较强的抗肿瘤免疫。
【Abstract】 The application of cartilage polysaccharide(PS) in preparation of tumor vaccine was studied in this paper.The conditions for preparation of the tumor vaccine were optimized using animal models.Mice immune organs and proliferation of spleen lymphocytes were measured and the changes of L1210 cell protein were detected through SDS-PAGE.Result showed that PS & cell 48 h had good immunity effects,protect the immune organs and obviously increase mice humoral immunity and also stimulate cellular immunity,detect specific antibody through cellular immunofluorescence.It inferred that changes of cellular protein induced the anti-tumor immune effects.
