盐酸戊乙奎醚对兔脑血管痉挛损伤的影响

【摘要】 目的:研究盐酸戊乙奎醚对蛛网膜下腔出血后脑血管痉挛的影响。方法:40只成年新西兰大白兔随机分对照组(NS组),模型组(SAH组),枕大池药物(SAH+CH组)组及治疗对照组(SAH+Vehicle组);动物用改良后单次注血法造模,所有动物在48 h后处死,取材计算基底动脉血管皱缩指数(CC),光镜下观察血管内膜痉挛的情况,脑脊液测定超氧化物歧化酶(SOD)、丙二醛(MDA)、S-100β蛋白(S-100β)。结果:改良动物模型短程、平稳、安全,光镜下观察药物组脑血管痉挛程度明显减轻;与SAH组比较,SAH+CM组CC值减小,P<0.05;与SAH组相比,SAH+CM组S-100β蛋白浓度明显降低,P<0.05;其余各组与NS组比较,SOD活性降低(P<0.01),MDA含量升高(P<0.01)。结论:盐酸戊乙奎醚可以缓解蛛网膜下腔出血后的脑血管痉挛和神经损伤。

【Abstract】 Objective: To investigate the effect of penehycldine hydrochloride(PHC) on cerebral vasospasm caused by subarachnoid hemorrhage(SAH),and to observe the stability of modifyed animal model of rabbit.Methods: A total of 40 adult male New-Zealand rabbits weighing 2.0~2.5 kg were randomly divided into 4 groups(n=10):group NS(normal control),group SAH(SAH model),group SP(SAH model treated by PHC),and group SV(SAH model injected with NS).SAH model was made by injection of autologous blood into the cisterna magna.Rabbits in group C were injected with saline into cisterna magna instead.SAH models in group SP were injected with PHC(0.12 mg/kg) in 30 min later and repeated once on the next day,and rabbits in group SV received twice injection of NS.All the animals were killed in 48 hours.The endangium spasm was observed under light microscope,corrugation coefficients(CC) on basilar artery were investigated using computerized image analysis,and SOD,MDA,and S~100β protein in cerebrospinal fluid were determined.Result:(1) The SAH animal model was established successfully;(2) CC values and S~100β protein contents were significantly lower in group SP than those in groups SAH and group SV(P<0.05);Compared with those in group NS,SOD activity decreased(P<0.01),and MDA contents increased(P<0.01) in other groups.Conclusion: Penehycldine hydrochloride could prevent cerebral vasospasm and injury of brain after SAH;The modifyed method for making rabbit vasospasm model is appropriate.
