【摘要】 目的:探讨口服Ⅱ型胶原(CⅡ)对大鼠佐剂型关节炎(AA)的治疗作用。方法:建立大鼠AA动物模型,观察口服高、中、低剂量的CⅡ后大鼠关节病变情况,流式细胞仪检测大鼠外周血T细胞亚群的变化,原位杂交法检测大鼠脾脏和派氏淋巴结(Peyerpatchespp)中转化生长因子β(TGF-β)、IL-12水平的变化。结果:口服高、中、低剂量的Ⅱ型胶原均可减轻关节病变的发生,高剂量组作用最显著,大鼠外周血CD4+T细胞和CD8+T细胞水平与模型组比较均有显著性差异,脾脏和pp淋巴结TGF-β水平较模型组升高,IL-12无明显改变,pp淋巴结TGF-β阳性细胞数高于脾脏。结论:TGF-β是口服耐受抑制AA的重要细胞因子,肠道黏膜免疫系统在口服耐受的诱导中可能发挥重要作用。更多还原

【Abstract】 Objective:To investigate the effects of oral administration of collagen(CⅡ)on TGF-β and IL-12 in rats with adjuvant arthritis(AA).Methods:Rat AA model was established by immunization with Freund’s adjuvant,and received different doses of CⅡ orally during a 2-week period afterwards.Incidence and severity of arthritis were assessed by calculation of mean arthritis index.T cell subsets in peripheral blood were assayed by immunofluorescence technique and concentrations of TGF-β,IL-12 in spleen and Peyer patches(pp)of rats were determined with in situ hybridization kit separately.Results:CⅡ at different doses could ameliorate the adjuvant induced arthritis,especially at high dose.Compared with control group,T cell subset CD4 decreased while T cell subset CD8 increased significantly when treated with high dose of CⅡ,and TGF-β concentration also increased significantly,especially in pp.No change of IL-12 content was found.There were more TGF-β positive cells in pp than in spleen.Conclusions:TGF-β is one of the important inhibitory cytokines to AA with cell tolerance,and the immune system in intestinal mucosa may play an important role in suppression of AA by oral tolerence.更多还原
