单唾液酸神经节苷脂对体外循环后大鼠脑组织Na~+-K~+-ATP酶活性的影响

【摘要】 目的:探讨单唾液酸神经节苷脂(GM1)对大鼠体外循环(CPB)后脑组织的含水量和Na+-K+-ATP酶活性的影响。方法:选用雄性SD大鼠,经右颈静脉插管引流,尾动脉插管灌注建立CPB,转流时间1h,建立CPB动物模型。随机分为CPB模型组、CPB模型+GM1组、输血组和假手术组,每组10只。用干湿法测定脑组织含水量,定磷法测定脑组织的Na+-K+-ATP酶活性。结果:CPB24h后,CPB模型组和CPB模型+GM1组大鼠脑组织含水量均较假手术组明显增加(P<0.01),但CPB模型+GM1组大鼠脑组织含水量较CPB模型组明显减少(P<0.05);CPB模型组脑组织中的Na+-K+-ATP酶活性由正常的[(17.03±4.17)mol·Pi/g·protein·h-1]下降到[(12.45±2.64)mol·Pi/g·protein·h-1](P<0.01),CPB模型+GM1组下降到[(14.72±3.29)mol·Pi/g·protein·h-1](P<0.05),与CPB模型组比较差异有统计学意义(P<0.05)。结论:CPB可抑制Na+-K+-ATP酶活性,使脑水肿加重;GM1可起到稳定细胞膜Na+-K+-ATP酶的作用。

【Abstract】 Objective:To observe the effect of monosialoteterahexosyl ganglioside 1(GM1)on the activity of Na+-K+-ATPase of brain tissues after cardiopulmonary bypass(CPB)in rats.Methods:SPF-class adult male SD rats were randomly divided into CPB group,CPB+ GM1group,transfusion group and shamoprated group.After establishing the rat model of CPB,GM1with 20mg/kg was added to the priming solution.All rats were anesthetized to collect the whole cerebral tissues for detecting the activity of Na+-K+ATPase and water content after 24hours.Results:The activity of Na+-K+-ATPase in cerebral tissues was descended from the normal(17.03±4.17)mol·Pi/g protein·h-1 to(12.45±2.64)mol·Pi/g protein· h-1.GM1administration obviously enhanced the activity of Na+-K+-ATPase to(14.72±3.29)mol·Pi/g protein·h-1.GM1can stabilize the functional activity of Na+-K+-ATPase of neuronal cytomembrane,and moderate cerebral edema from the brain injuries associated with CPB.Conclusion:CPB inhibits the Na+K+-ATP activity and increases brain edema.GM1can stabilize the functional activity of Na+-K+-ATPase of neuronal cytomembrane,and moderate cerebral edema from the brain injuries associated with CPB.
