近临界水中禽类废弃物超低酸水解加工制备氨基酸的工艺优化
Optimization for Amino Acids Production Technology from Bird′s Waste by Hydrolysis at Extremely Dilute Acids in Subcritical Water
【摘要】 为寻求禽类废弃物的有效利用途径,对其进行在近临界水中超低酸水解加工制备氨基酸的工艺优化。用AAA-Direct氨基酸分析仪对水解产物中的氨基酸进行定性和定量分析。以鸡肠为原料,氨基酸总收率为指标,讨论了反应温度、反应时间、硫酸浓度对氨基酸总收率的影响,并通过正交试验确定了鸡肠近临界超低酸水解的较佳工艺条件。实验结果表明,鸡肠水解后可得17种氨基酸。近临界超低酸水解法高效、工艺简单、对环境友好,在较佳的工艺条件下:T=533K,t=28min,H2SO4=0.02%,氨基酸的总收率可达11.49%。
【Abstract】 In order to look for the effective way of bird’s waste utilization, the optimization of bird’s waste hydrolysis technology for amino acid production by using extremely dilute acid in subcritical water was conducted. The quality and quantity analysis of amino acid obtained during hydrolysis were carried out by AAA-Direct analyzer (Biological Liquid Chromatography). The experiments take chicken intestines as the raw material and the yield of amino acid as the target. The effects of reaction temperature and time, and the H2SO4 concentration on the yield of amino acid were investigated. The optimum hydrolysis conditions for the chicken intestines were studied by the orthogonal tests. The results show that 17 kinds of amino acids can be obtained, and the technology of amino acid production via bird’s waste hydrolysis by extremely dilute acid in subcritical water has the advantages of high efficiency, simple process and being friendly to environment. The optimum hydrolysis conditions for amino acids production from bird’s waste were found as follows: reaction temperature of 533 K, reaction time of 28 min and H2SO4 concentration of 0.02%. Under above conditions, the yield of amino acids can reach 11.4%.
