AB-8大孔吸附树脂对稻壳黄酮的分离纯化工艺研究
Study on the separation and purification technology for total flavonoids from paddy hull by AB-8 macroporous adsorption resin
【摘要】 以对稻壳黄酮的吸附率、解吸率为指标,采用正交试验研究了AB-8大孔吸附树脂对稻壳中总黄酮的分离纯化工艺,分析了上柱及解吸液浓度、pH和流速对动态吸附、解吸的影响.结果表明,AB-8树脂对稻壳中黄酮类化合物的分离纯化优化条件为:上柱液浓度为1.447mg·mL-1,pH5.0,流速1mL·min-1;乙醇解吸液体积分数50%,pH6.0,流速为1mL·min-1.此外,通过动态吸附泄漏曲线和动态解吸曲线确定的上柱液与解吸液体积均为5.5倍树脂柱体积.
【Abstract】 Taking the adsorption and desorption rate of paddy hull flavonoids as indexes,the orthogonal test was performed to investigate the separation and purification technology for total flavonoids from paddy hull by AB-8 macroporous adsorption resin.The effects of loading solution and eluant concentration,pH and the flow velocity on the dynamic adsorption and desorption were also searched.The results showed that AB-8 resin had good property in purification and separation,and its optimal separation and purification technological conditions were as follows:the concentration of loading solution 1.447mg· mL-1 pH 5.0;ethanol eluant 50%pH 6.0,and the velocity of them were both 1mL·min-1.Besides,the leaking curve of dynamic adsorption and the dynamic desorption curve were examined,and the appropriate volume of loading solution and the eluant volume were both 5.5 times of resin volume.
