IGF-1对高糖诱导的小鼠早期胚胎凋亡的抑制作用

【摘要】 目的探讨胰岛素样生长因子1(IGF-1)对体外培养小鼠囊胚细胞凋亡的抑制作用。方法获取妊娠3.5d小鼠囊胚,分别移入3个培养皿中,分别为A、B、C三组:A组(基础培养液);B组(基础培养液+30mmol/L的葡萄糖溶液);C组(基础培养液+30mmol/L的葡萄糖溶液+100ng/ml的人重组IGF-1)。连续培养72h后,采用免疫组化S-P法检测各组囊胚细胞中Bax和Fas的表达,利用计算机图像分析技术测量各组囊胚细胞中Bax和Fas蛋白表达的平均光密度和平均阳性面积。结果B组囊胚细胞中Bax和Fas的表达明显高于A组和C组(P<0.05)结论IGF-1对高糖诱导的小鼠着床前早期胚胎凋亡起了抑制作用。

【Abstract】 Objective To observe the anti-apoptotic effect of insulin-like growth factor 1 (IGF-1) on blastocyst cells in vitro. Methods Mouse blastocysts of 3.5d were put respectively into 3 culture dishes: group A(basal culture solution), group B(basal culture solution + 30mmol/L glucose),and group C (basal culture solution + 30mmol/L glucose + 100ng/ml IGF-1). The expressions of Bax and Fas in each group were detected by immunohistochemical S-P method after continuous cultivation for 72 hours. We used an image analysis system to measure the average optical density and the area of Bax and Fas expressions in each group . Results The expressions of Bax and Fas were significantly higher in group B than in group A and group C (P<0.05). Conclusion IGF-1 plays an inhibitory role in early-period embryonic apoptosis induced by high glucose.
