拟南芥psrp-3基因的PCR扩增与生物信息学分析

【摘要】 采用PCR法获得拟南芥叶绿体核糖体psrp-3基因序列并进行生物信息学分析.根据psrp-3基因序列保守区设计2对引物,通过RT-PCR获得预期大小的基因序列,琼脂糖凝胶电泳鉴定目的片段.利用生物信息学软件对拟南芥psrp-3基因序列进行同源比对、氨基酸组成、功能域、二级结构、疏水性、蛋白质功能及系统进化分析和预测.结果表明,RT-PCR获得了一段753 bp的序列,psrp-3蛋白是等电点为9.27的亲水性稳定蛋白,包含一个结构域和一个区域,α螺旋和不规则卷曲是其蛋白质二级结构的主要结构元件,β折叠和伸展链散布其中,和菠菜、葡萄、蓖麻、大豆等的psrp-3蛋白有较高的同源性.为进一步了解psrp-3基因的功能和作用机制打下基础.

【Abstract】 The psrp-3 gene was amplified from arabidopsis thaliana by PCR,with primers designed according to public psrp-3 sequences.We analyzed the conservative functional domains,protein secondary structure,and their evolutionary relationships among psrp-3 gene of arabidopsis thaliana and other nineteen species by using approaches of bioinformatics.The open reading frame of psrp-3 gene has 753 bp in length.The estimated isoelectric point of the putative protein was 9.27.Hydrophobic analysis indicted that psrp-3 was a hydrophilic protein.The psrp-3 generally contains one functional domain.The predicted secondary structure of the protein had mainly about α-helix and random coil,also had β-turn and extended strand among them.It shared high homologies in amino acid sequence level with spinacia oleracea,vitis vinifera,ricinus communis,and glycine max,respectively.PCR amplification and bioinformatics analysis of psrp-3 gene in arabidopsis thaliana will be helpful for the further analysis of psrp-3 protein.
