固相萃取-高效液相色谱法检测牛奶和猪肉中5种β-内酰胺类抗生素
Development of SPE-HPLC method for simultaneous determination of five beta-lactam antibiotic residues in milk and swine meat samples
【摘要】 建立了牛奶和猪肉样品中氨苄西林、青霉素V、青霉素G、苯唑西林和氯唑西林等5种β-内酰胺类抗生素残留量的高效液相色谱(HPLC)测定方法。实验采用固相萃取法(SPE)富集抗生素残留,考察了样品前处理方法;并对色谱分离条件加以优化,在C18色谱柱上以乙腈-0.1%氨水进行梯度洗脱,可在10min内实现5种抗生素的分离与检测,检测波长为210nm。方法可用于动物性食品中上述抗生素残留的同时检测。
【Abstract】 A rapid high performance liquid chromatographic (HPLC) method with UV detection was developed for simultaneous determination of five beta-lactam antibiotics:ampicilin;penicillin G;penicillin V;cloxacillin and oxacillin in milk and swine meat.A chromatographic separation was achieved in 10 min on a C18 column with the mobile phase consisting of acetonitrile and 0.1% ammonia mixture in a gradient mode.Detection was carried out at 210 nm.Validation experiments were performed to demonstrate linear ranges;precision,recovery and limit of detection (LOD).The principal advantages of this method are simplicity,quickness,and no derivatization being needed.
