新型脱氧核糖核酸光纤生物传感器的研制及其性能评价

【摘要】 研制了一种新型脱氧核糖核酸(DNA)光纤生物传感器,以带尾纤的脉冲半导体激光器为激发光源,结合流动进样系统,能够快速方便地对光纤传感器表面发生的DNA杂交反应进行测定。研究表明:通过链霉亲和素/生物素作用修饰到传感器表面的DNA探针对Dylight680荧光标记的完全互补DNA有良好的响应,而对非互补的DNA基本没有响应。系统检测灵敏度可达到4.5×10-10mol/L;线性范围为1×10-9~3×10-8mol/L。光纤传感器具有良好的再生性能。

【Abstract】 A novel DNA fiber optic biosensor has been successfully developed.A pulse diode laser with pigtail was used.Integrated with flow-injection system,the sensor could detect DNA hybridization on the fiber optic sensor surface rapidly and conveniently.The detection principle,system structure,modification of fiber optic sensor,and DNA hybridization were discussed.The experimental results showed that the biotinylated DNA probe immobilized on fiber sensor via biotin-streptavidin interactions could greatly hybridizate with full-matched DNA labeled by Dylight 680 but not with mis-matched DNA labeled by Dylight 680.The sensitivity of detection was 4.5×10-10 mol/L and a linear response range was observed from 1×10-9 to 3×10-8 mol/L.The sensor surface could be regenerated without obvious loss of hybridization activity.The new DNA biosensor showed enormous potential for the detection of clinical,diagnostic and environmental samples.
