核因子NF-κB在胰腺炎细胞模型中的实验研究

【摘要】 目的:探讨急性胰腺炎(AP)时核因子NF-κB激活的情况及NF-κB在AP病程中的作用。方法:实验分为两组:A组(对照组),B组(将AR42J细胞以caerulein作用制成AP细胞模型)。检测淀粉酶及乳酸脱氢酶(LDH)释放的情况;另取1ml培养液上清加入接种巨噬细胞的培养板中培养,然后,收集巨噬细胞进行NF-κB激活检测。结果:B组胰腺腺泡细胞内容物淀粉酶、LDH的释放与A组比较明显增加(P<0.05);B组NF-κB激活较A组明显升高(P<0.05)。结论:AP时可影响核因子NF-κB激活,胰腺炎胰腺腺泡细胞内容物的释放增加时则NF-κB激活水平升高。

【Abstract】 Objective To investigate the enhancememnt of NF-κB activation with pancreatitis cell models.Methods Models of pancreatitis cells were prepared with pancreatic alveolar cell line AR42J cells stimulated with caerulein.Amylase and LDH contents in the culture supernatant were determined with colorimetry.Peritoneal macrophages were retreived from Wistar rats,cultured with the model pancreatitis cells and NF-κB activity examined with flow-cytometry.Untreated AR42J cell cultures served as controls.Results The release of amylase and LDH in models were significantly increased(vs controls,P<0.05).The cytokine NF-κB activity was significantly higher in macrophage calture treated with model cells(P<0.05).Conclusion The release of amylase and LDH in pancreatitis model cells could enhance the NF-κB activation of macrophages.
