超分割放疗对宫颈癌组织中细胞增殖凋亡相关基因mRNA表达的影响
The effects of hyperfractionated radiotherapy on expression of proliferation nuclear antigen and apoptosis-associated genes in cervical carcinoma tissue
【摘要】 目的探讨超分割放疗对宫颈癌组织中增殖核抗原(PCNA)、细胞凋亡相关基因bcl-2、bax mRNA表达的影响。方法用RT-PCR法检测超分割组23例,常规分割组10例宫颈癌组织在放疗前、后及放疗中PCNA、bcl-2和bax基因表达的情况、动态变化和差异。结果放疗2周后较放疗前超分割组bax基因的表达明显增强(P<0.05),常规分割组则差异无统计学意义(P>0.05)。PCNA、bcl-2基因的表达变化差异无统计学意义(P>0.05)。结论超分割放疗比常规分割放疗更好的上调bax基因的表达,增强诱导肿瘤细胞凋亡。
【Abstract】 Objective To discuss the effects of hyperfractionated radiotherapy on expression of cervical carcinoma cells proliferation nuclear antigen (PCNA), apoptosis-related gene bcl-2 and bax.Methods Cervical cancer tissues were obtained from 23 patients who received hyperfractionated radiotherapy (hyperfractionated group) and 10 patients who underwent conventional fractionated radiotherapy (conventional fractionated group).Before and in the course of treatment (after 2 weeks and 4 weeks) , the expression of PCNA,bcl-2 and bax genes in both groups were determined by RT-PCR method.Results In hyperfractionated group, expression of bax mRNA was significantly increased after 2 weeks treatment compared to that before treatment, while in conventional fractionated group, no significance difference was found. There were no significance difference on expression of bcl-2 and PCNA mRNA pre- and post-radiotherapy in both groups.Conclusion Hyperfraction radiotherapy could up-regulate the expression of bax and thus induct apoptosis.
