基因芯片研究小金丹对人肝癌细胞信号转导基因表达的影响
Effect of Xiao Jin Dan on Expression of Signal Transduction Genes in SMMC-7721 Cell line by DNA Microarray
【摘要】 目的观察中药复方小金丹对人肝癌细胞信号转导基因表达的影响。方法制备小金丹含药血清,用不同浓度的小金丹含药血清处理人SMMC-7721细胞。采用基因芯片技术检测小金丹含药血清处理后人肝癌细胞SMMC-7721细胞信号转导基因表达变化。结果小金丹处理SMMC-7721细胞后,所检测的96个信号转导基因中,有77个基因的表达出现显著上调或下调(比值<0.5或>2.0)。剂量不同,其影响的基因种类和程度也有所不同。结论小金丹可显著影响人肝癌细胞信号转导基因的表达。
【Abstract】 Objective To observe the effect of Xiao Jin Dan(XJD) on the expression of signal transduction genes in SMMC-7721 cells.Methods Serum containing XJD was prepared in SD rats.The expression of signal transduction genes in SMMC-7721 cell before or after serum containing XJD treatment were detected by microarray.Results Of 96 signal transduction genes detected,77 showed significant difference in expression after treatment with XJD,compared with control group(Ratio<0.5,or>2.0).Kind and intension of gene expression were different among various dose of XJD treatment groups.Conclusion XJD affects significantly expression of signal transduction genes.
