人工养殖鲶的TRAP遗传分析
Genetic analysis of cultured catfish Silurus asotus using TRAP technique
【摘要】 建立了鲶Silurus asotus的TRAP-PCR反应体系,采用24个TRAP引物组合对鲶人工选育群体进行引物适用性的研究,将获得的多态性位点用于研究鲶群体的遗传结构。结果表明:有9个引物组合可产生有效位点,其中多态引物占所筛选引物总数的37.5%,平均每条引物扩增出15个位点。共扩增出135个位点,多态位点数为75,多态位点比例为55.56%。说明TRAP标记技术适用于鲶的遗传多样性研究,可以作为构建鲶遗传连锁图谱的分子标记。
【Abstract】 TRAP(Target Region Amplified Polymorphism) was first used for research of genetic diversity in catfish Silurus asotus.Nine pairs of primers selected from 24 primers produced good amplified patterns with 37.5% of polymorphic primer was in all amplified primers.There were 15 loci amplified by one primer,and 75 polymorphic loci were obtained in all 135 amplified loci with 55.56% of polymorphic loci.The genetic variation analyses showed that TRAP marker can be used in genetic structure research of the catfish.The higher percentage of polymorphic primer number indicated that TRAP as a DNA molecular marker can be used in genetic linkage map research for Silurus asotus.
