几种理化因子对厚壳贻贝精子活力的影响
The influence of physical-chemical factors upon Mytilus coruscus sperm motility
【摘要】 通过盐度、pH值及海水主要离子对厚壳贻贝Mytilus coruscus精子的激活效果实验研究结果表明,盐度为5~35的溶液对厚壳贻贝精子都可激活,但精子激活率和运动时间均存在差异,在盐度为20的溶液中,精子激活率最高,为63.7%,精子运动时间最长,为37.5 min;高盐度环境下的精子运动时间和激活率长于、高于低盐度环境下的精子运动时间和激活率。不同pH值的海水对厚壳贻贝精子运动时间和激活率的影响也存在差异,在pH值为7.0~8.5的海水中,厚壳贻贝精子的激活率和运动时间随着pH值的增加而上升;在pH值为8.5~9.5的海水中,精子激活率和运动时间随着pH值的增加而下降;在pH值为8.5的海水中,精子运动时间最长,为54 min左右,精子激活率最高,为67.4%;高pH值海水中精子激活率和运动时间均比低pH值海水中的高。质量分数为1%~5%的NH4Cl和CaCl2溶液对厚壳贻贝精子没有激活作用。质量分数为1%和2%的KNO3溶液不能激活厚壳贻贝精子,质量分数为3%~5%的KNO3溶液对精子能起到激活作用,且激活作用随着溶液质量分数的增加而提高。质量分数为1%~4%的MgCl2溶液对厚壳贻贝精子的激活作用不明显,只有在质量分数为5%时该溶液才能激活少许精子。除质量分数为1%和5%的KCl溶液不能激活厚壳贻贝精子外,质量分数为2%~4%的该溶液均能激活厚壳贻贝精子,质量分数为3%时该溶液对厚壳贻贝精子的激活率最高。质量分数为1%~3%的NaCl溶液能激活厚壳贻贝精子,而质量分数为4%和5%的NaCl溶液不能激活厚壳贻贝精子。Na+、K+和Mg2+溶液均能改变厚壳贻贝精子细胞膜的通透性,对该精子起到激活作用。
【Abstract】 The impacts of pH,salinity and main ions in seawater on the sperm motility and moving-time of Mytilus coruscus are studied.The results show that the sperm of the Mytilus coruscus could be activation and survive for a long time when the salinity is from 5 to 35,but the activation rate and moving-time are different and the most suitable salinity is 20 with the highest sperm activation rate of 63.7% and the longest moving-time of 37.5 min.The lifetime and activation rate in high salinity is higher than that in low salinity.Experiments also show that the sperm motility and moving-time changed with pH of water.When pH was 7.0~8.5,the activation rate and moving-time of sperm gradually increased.When pH was 8.5 to 9.5,the activation rate and moving-time gradually declined.When pH was 8.5,the moving-time was the longest and the activation rate highest,which were about 54 min and 67.4%.The lifetime and activation rate in high pH is higher than that in low pH.NH4Cl and CaCl2 solution of 1% to 5% concentration and KNO3 solution of 1% and 2% concentration had no activation to the sperm.KNO3 solution of 3% to 5% concentration had activation on the sperm,and the activation effect increased with the increase of the concentration.The activation of MgCl2 solution of 1% to 4% concentration on the sperm wasn’t evident,only when the concentration was up to 5%,a small quantity of sperms was activated.Except the concentration of 1% and 5%,KCl solution of 2% to 4% concentration could activate the sperm,and the activation rate was the highest when the concentration was 3%.NaCl solution of 1% to 3% concentration can activate the sperm,while that of 4% and 5% concentration can not.That is to say,Na+,K+ and Mg2+ solution can alter the cell membrane permeability of the sperm and has activation on the sperm.
