改良型序列特异性引物PCR用于脂联素基因单核苷酸多态性的检测
Application of Improved SSP-PCR in Detection for Single Nucleotide Polymorphism
【摘要】 目的运用序列特异性引物PCR技术(SSP-PCR)研究基因单核苷酸多态性。方法根据GenBank中人脂联素基因(APM1)序列及SNP信息,利用Primer5.0设计引物,通过PCR技术检测APM1的SNPs。结果建立并优化了序列特异性引物PCR(SSP-PCR)技术,并可快速、直观、准确地进行基因分型。结论改良的SSP-PCR技术是一种特异性更高、操作简便的SNP检测方法。
【Abstract】 Objective To research single nucleotide polymorphism(SNP) of genes by sequence-specific-primers polymerase chain reaction(SSP-PCR). Methods According to information of adiponectin gene (APMI) of human from GenBank, specific primers were designed with Primer 5.0, and SNPs of APMI were detected by PCR. Results SSP-PCR was successfully established and improved. It could carry out genotyping rapidly, directly and exactly. Conclusions Improved SSP-PCR technique is a more specific and easier way to detect gene polymorphism.
