栀子苷对大鼠局灶性脑缺血模型缺血脑组织HIF-1α蛋白和RTP801的影响

【摘要】 目的观察栀子苷对脑缺血再灌住模型大鼠海马组织HIF-1α,HIF-1α依赖的凋亡相关基因RTP801 mRNA表达的影响。方法实验动物随机分为假手术组、模型组、栀子苷低剂量组、栀子苷中剂量组、栀子苷高剂量组、尼莫地平组。制备大鼠局灶性脑缺血模型,以栀子苷、尼莫地平腹腔注射给药,免疫组化法观察海马组织HIF-1α的表达情况,RT-PCR检测RTP801 mRNA表达的情况。结果栀子苷治疗组和尼莫地平组大鼠海马组织HIF-1α的表达均降低,RTP801 mRNA的表达也均下降。结论栀子苷抑制局灶性脑缺血损伤引起的的HIF-1α和HIF-1依赖性凋亡相关基因RTP801 mRNA的表达,从而减少神经元凋亡。更多还原

【Abstract】 Objective To investigate the influence of gardnoside on the expression of HIF-1αand RTP801 mRNA in rats with focal cerebral ischemia.Methods Experimental rats were randomly divided into six groups:sham groups,modle group,low-dose group with gardnoside,average-dose group with gardnoside,high-dose group with gardnoside,and nimodipine-group.Rats were prepared for models of focal cerebral ischemia.Drugs were administered by enterocolia.The Immunohistochemical steining showed the expression of HIF-1α.RT-PCR showed the expression of RTP801 mRNA.Results The expression HIF-1αand RTP801 mRNA was lower compared with the model group.Conclusion Gardnoside could restrain the expression HIF-1αand RTP801 mRNA to reduce neurological cell apoptosis.更多还原
