豫杂一号泡桐体细胞同源四倍体诱导及其体外植株再生
Induction of Autotetraploid from the Somatic Cell of Paulownia tomentosa×P.fortunei and Its in Vitro Plantlet Regeneration
【摘要】 在含不同质量浓度秋水仙素固液双层培养基上,以叶片为外植体进行了豫杂一号泡桐同源四倍体的诱导,同时建立了其体外植株再生系统。结果表明,秋水仙素质量浓度和处理时间对豫杂一号泡桐同源四倍体诱导率影响显著,外植体预培养时间影响不显著。在含20 mg.L-1秋水仙素MS培养基上处理48 h,未经预培养叶片四倍体诱导率最高达21.2%。同源四倍体幼苗叶片及其单气孔器较二倍体变大,气孔密度变小,叶绿素含量和SOD活性升高,MDA含量和POD活性降低。此外,叶片是同源四倍体豫杂一号泡桐体外植株再生的最佳外植体,MS+0.5 mg.L-1NAA+16 mg.L-1BA和1/2MS+0.1 mg.L-1NAA分别是其芽诱导和根诱导的最适培养基。
【Abstract】 A study was conducted to induce autotetraploid from diploidy leaves of Paulownia tomentosa × P.fortunei on the liquid-solid double layer MS media.The in vitro plantlet regeneration was established using different types of explants.Results indicate that the concentrations of colchicine and the treatment period have significant influences on the induction rate of the autotetraploidy while pre-culture time has a insignificant effect on induction rate.The highest induction rate reached 21.2% from non-precultured leaves treated with 20.0 mg·L-1 colchicine for 48.0 h on the MS medium.Leaves of the autotetraploid were larger and thicker,and the stomatal size and density on the leaf were larger and thinner than those of the diploid.The chlorophyll content and SOD activity of the autotetraploid were higher while MDA content and POD activity were lower than those of the diploids.Moreover,leaves are the suitable explants for shoot induction in the the system of in vitro plantlet regeneration of the autotetraploids.MS medium supplemented with 0.5 mg·L-1 NAA and 16.0 mg·L-1 BA and 12 MS with 0.1 mg·L-1 NAA were the optimal media for shoot and root induction respectively.
