辽宁地区主栽杨树品种的SRAP标记遗传多样性分析

【摘要】 采用SRAP分子标记技术对17个采自辽宁不同地区的杨树材料进行了遗传多样性研究。结果表明:利用31对SRAP引物共扩增出685个多态性条带,平均每个引物扩增出23.2条带,多态性比率为95.3%,各品种之间的遗传相似系数在0.3622~0.9475,平均相似系数为0.6549。通过UPGMA聚类分析将17个杨树材料分为4个群,其划分结果与传统分类基本一致,证明SRAP标记是一种适于杨树遗传多样性研究的分子标记技术。聚类结果表明,品种间遗传差异最小是银中杨和毛白杨,差异最大的是意大利45杨和毛白杨;2个小胡杨的种内遗传差异最小,而2个意大利45杨的种内遗传差异较大。

【Abstract】 Sequence related amplified polymorphism(SRAP)markers were used to analyse the genetic relationships of 17 Populus materials,collected in Liaoning area.A total of 685 polymorphic bands were amplified using 31 primers.Each primer could amplify 23.2 bands on average,and the average percentage of polymorphic bands was 95.3%.The genetic similarity coefficients among the tested cultivars ranged from 0.3622 to 0.9475,averaging 0.6549.The cluster analysis showed that these materials could be divided into four main groups by using the UPGMA.The clustering result was generally consistent with the previous classification based on conventional methods,which implies that SRAP marker is a useful method for evaluating genetic diversities of Populus.The clustering results also show that there is the smallest genetic difference between P.alba and P.tomentosa,whereas the largest difference between P.×canadensis’I-45’ and P.tomentosa,and there is the largest genetic intraspecies variation in P.×canadensis ’I-45’ while the lowest one in P.simonii×P.euphratica.
