基于COI基因限制性内切酶谱差异对几种杨树蛀干害虫幼虫的辅助鉴别
Assistant Identification of Several Species of Poplar Borer Larva Based on Different Restriction Enzyme Characteristic in Their COI Genes
【摘要】 以黑龙江省杨树上的4种主要蛀干害虫:杨干象、青杨楔天牛、青杨脊虎天牛和光肩星天牛幼虫为研究对象,使用PCR方法体外扩增并得到了4种幼虫mtDNA中的细胞色素C氧化酶I(COI)基因片段,并进行了序列分析。基于扩增得到的这4种害虫COI基因的限制性内切酶酶谱的比较,可筛选出特异性的内切酶,并通过酶切方法对这4种害虫各自的COI基因进行了特异性鉴别、区分。在此类危险性害虫的检验检疫工作中,通过形态学及危害状进行初步判断的基础上,利用该方法进一步进行辅助鉴别,可大大提高杨树蛀干害虫幼虫检疫、鉴定的准确性。
【Abstract】 DNA fragments of COI gene of larvae of Cryptorrhynchus lapathi (Linnaeus),Saperda populnea (Linnaeus),Xylotrechus rusticus (Linnaeus),and Anoplophora glabripennis (Motschulsky),the main poplar stem-boring pests in Heilongjiang Province,were amplified by PCR,and then sequenced and analyzed. Maps of COI gene restriction enzyme of the four species of pests were obtained. Moreover,three restriction enzymes were selected to cut the amplified COI gene fragments of X. rusticus,C. lapathi,and A. glabripennis specifically,but the COI gene of S. populnea could not be cut by any of these three restriction enzymes. Therefore,the assistant method could be applied to the inspection and quarantine of these kinds of pests,based on the primary identification of pests by their morphological characters and damage features,which would improve the identification accuracy.
