【摘要】 目的:研究慢性乙型肝炎(CHB)患者进行核苷(酸)类似物抗病毒治疗后,不同疗效患者树突状细胞(dendritic cell,DC)功能的差异。方法:选取56例HBeAg阳性的慢性乙型肝炎(CHB)患者用拉米夫定与阿德福韦酯交替治疗104周后,依据患者HBeAg/HbeAb血清学转换情况,分为转换组19例和未转换组37例,分别抽取外周血分离和培养DC,流式细胞仪检测和比较转换组和未转换组DC表面分子HLA-DR、CD80和CD86表达水平;用MTT法测定DC刺激混合淋巴细胞反应能力,比较两组的DC刺激指数(SI)。结果:(1)所有检测对象的DC存活率无统计学差异(P>0.05);(2)转换组的DC表面分子HLA-DR、CD80的表达水平较未转换组明显增加(P<0.05);CD86表达水平略高于未转换组但无显著统计学差异(P>0.05);(3)转换组较未转换组的SI显著增加,有统计学差异(P<0.05)。结论:核苷(酸)类似物抗病毒治疗CHB增强DC功能可能与患者HBeAg血清学转换有关。更多还原

【Abstract】 Objective:To investigate the function of dendritic cell(DC) from different effectiveness of patients with chronic hepatitis B by antiviral therapy with nucleoside(acid) analogues.Methods:Fifty-six CHB patients with HBeAg-positive were included in this study.All patients were treated with nucleoside(acid) analogues(lamivadine and adefovir alternation therapy for 104 weeks),The patients were divided into two groups according to their HBeAg seroconversion: HBeAg-positive and HBeAb-positive.Then the patients’ peripheral blood were separated to culture DC.The phenotypes of DC,such as HLA-DR、CD80、CD86,were subjected to flow cytometric analysis.The mixed lymphocyte stimulate reaction capacity of DC were detected by MTT,in order to calculate index(SI).Then the function of DC were compared between HBeAg seroconversion and without seroconversion after therapy.Results:(1)Survival rates of DC from all patients had no statistical difference(P<0.05);(2)The expression of costimulating molecules CD80、HLA-DR of DC from patients with seroconversion is higher than that of without seroconversion.The expression of CD86 was increasing,but it had no significantly statistical difference(P>0.05);(3)As compared with patients without seroconversion,SI of patients with seroconversion was increased markedly,there was significantly statistical difference between two groups(P<0.05).Conclusion: The function improvement of DC in CHB patients with nucleoside(acid) analogues antiviral therapy may be correlative with HBeAg seroconversion.更多还原
