【摘要】 目的通过检测系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)凋亡相关基因程序化细胞死亡因子5(PDCD5)mRNA表达变化,从而探讨PDCD5基因与SLE发生与发展的关系。方法应用逆转录PCR(RT-PCR)半定量检测67例SLE患者和50例健康者PBMC中PDCD5 mRNA的表达。结果 SLE活动期患者PDCD5mRNA阳性表达率明显高于健康者(P=0.00);SLE活动期患者PDCD5 mRNA表达水平明显高于稳定期患者和健康者(P<0.01),稳定期患者PDCD5 mRNA比健康者也明显升高(P<0.01)。SLE活动期患者PBMC中PDCD5的表达与反应SLE疾病活动的指标如SLEDAI、补体C3、抗ds-DNA抗体等呈显著相关(P<0.05)。狼疮性肾炎患者和非狼疮性肾炎患者PDCD5表达差异无统计学意义(P>0.05)。结论 SLE患者PBMC中PDCD5表达上调,且与SLE疾病活动相关,提示PDCD5通过增加表达量而导致异常淋巴细胞凋亡,参与SLE的发生、发展。

【Abstract】 Objective To study the expression of programmed cell death 5 gene(PDCD5) mRNA in peripheral blood mononuclear cells(PBMC) from Systemic lupus erythematosus(SLE) patients and investigate the role of PDCD5 in the occurrence and development of SLE.Methods The expression of PDCD5 mRNA in PBMC from 67 SLE patients and 50 healthy cases were detected by semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR).Results The positive rate of PDCD5 mRNA from active SLE patients was significantly higher than that in normal cases(P=0.00).The expression levels of PDCD5 mRNA in active SLE patients was significantly higher than that of inactive SLE patients and that of normal cases.PDCD5 mRNA expression levers of inactive SLE patients was significantly higher than that of normal cases(P<0.01).Significant correlation was found between the PDCD5 expression in PBMC from active SLE patients and clinical parameters such as SLEDAI,C3 and anti ds-DNA antibody(P>0.05).There was no significant difference in the expression of PDCD5 between lupus nephritis patients and non-lupus nephritis patients(P<0.05).Conclusion The up-regulation of expression of PDCD5 mRNA associated with disease activities in PBMC from SLE patients indicates that the abnormal apoptosis of lymphocytes resulted from the overexpression of PDCD5 perhaps plays an important role in the pathogenesis of human SLE.
