中药不同分期对实验性SD大鼠骨折愈合过程中成纤维细胞生长因子-2表达的作用

【摘要】 目的:研究中药不同分期对实验性SD大鼠骨折愈合过程中成纤维细胞生长因子-2(FGF-2)表达的影响,探讨中药治疗骨折的合理分期方法。方法:于SD大鼠双侧胫骨上1/3段前部的中点造成2mm×1.5mm的骨质缺损。222只雄性SD大鼠随机取6只作为对照组,其余于造模后随机平均分为4组:三期治疗组、二期治疗组、一期治疗组和模型组,分别给予相应干预措施。每组于术后不同时间点分别各处死6只,常规标本处理、组织学检查和免疫组化检测标本中的FGF-2,应用计算机图像分析系统进行定量分析。结果:①各组FGF-2表达平均阳性细胞数(PCN)及平均光密度(MOD):二期治疗组>三期治疗组>一期治疗组>模型组,P<0.05。②FGF-2表达高峰期:高峰期出现时间:二期、一期、三期治疗组和模型组依次推迟;高峰期持续时间:一期治疗组和二期治疗组,显著长于其它组。③FGF-2表达峰值:峰值出现时间:二期治疗组明显早于其它组,而其它各组间无明显差别;峰值水平:二期治疗组明显高于其它组(P<0.05);有三期治疗组>一期治疗组>模型组的趋势。结论:中药不同分期治疗均能有效促进SD大鼠骨折愈合过程中FGF-2的表达,其中二期治疗明显优于一期治疗和三期治疗,这可能是中药治疗骨折的最佳分期方法。

【Abstract】 Objective:To investigate the effect of stage treatment with TCM on FGF-2 expression in experimental fracture rats,to confirm effects and its modern medical mechanism of three-phase treatment for fracture with TCM,and to clarify reasonable phasing way in treatment for fracture with TCM.Methods:A 2-mm-wide1.5-mm-deep defect was bilaterally created as fracture healing model in the middle of the anterior part of the up-shaft of tibia of rats.At first,6 male SD rats were taken randomly into the normal control group.Postoperatively,222 male SD rats were divided randomly into four groups on average: three-phase,two-phase,and one-phase treatment group and model control group,and then every group was divided randomly into 9 small groups with 6 rats in each small group.Every group was respectively given corresponding interventions.After operation,at different time points,rats in a small group in each group were killed respectively.Specimens were prepared and examined histologically in routine.The expression of FGF-2 was detected by immunohistochemical staining and quantitatively analyzed by computer image analysis system.Results:①Mean MOD(mean optical density)of FGF-2 expression in every group:two-phase treatment group>three-phase treatment group>>one-phase treatment group>model control group(P<0.05).②Peak-stage of FGF-2 expression:occurring time of peak-stage in two-phase treatment group was obviously earlier than that in other groups.The lasting time of peak-stage in 2-phase treatment group was equal to that in 3-phase treatment group,and considerably longer than that in other groups.③The top level of peak of FGF-2 expression:occurring time of top level of peak in two-phase treatment group was obviously earlier than that in the other.Top level of peak in two-phase treatment group was significantly higher than that in the other(P<0.05).There was a tendency that top level in 3-phase treatment group was higher than that in 1-phase treatment group.④Change tendency of positive cell number of FGF-2 expression was generally consistent with that of MOD value.Conclusion: All of 3 kinds of different phasing treatment with TCM were able to effectively promote FGF-2 expression during fracture healing in SD rats.Of three ways,two-phase treatment was notablely stronger than the other in accelerating and raising FGF-2 expression.From this standpoint,it might be optimal phasing way in fracture treatment with TCM.
