大蝎子草乙酸乙酯部位对脂多糖诱导巨噬细胞炎症介质表达的影响

【摘要】 目的:研究大蝎子草乙酸乙酯部位对脂多糖(LPS)诱导大鼠腹腔巨噬细胞诱导型一氧化氮合酶(iNOS)表达、炎性因子一氧化氮(NO)与肿瘤坏死因子α(TNF-α)分泌的影响。方法:建立体外培养大鼠腹腔巨噬细胞,以LPS为致炎刺激剂,免疫细胞化学法研究大蝎子草乙酸乙酯部位对LPS诱导大鼠腹腔巨噬细胞iNOS表达的影响,Griess法分析培养液NO含量,间接MTT法检测培养液TNF-α含量。结果:LPS刺激大鼠腹腔巨噬细胞后,其iNOS表达显著增加,培养液中NO和TNF-α含量显著增加,大蝎子草乙酸乙酯部位对上述炎症介质的增加具有显著的抑制作用。结论:大蝎子草乙酸乙酯部位能抑制LPS诱导NO和TNF-α的产生,减少iNOS的表达,为其进一步研究开发奠定了实验基础。

【Abstract】 Objective:To investigate the inhibited effects of Herba Girardinia ethyl acetate extract on inducible nitric oxide synthase(iNOS)expression,and nitric oxide(NO)and tumor necrosis factor-alpha(TNF-α)excretion in rat peritoneal macrophages induced by lipopolysaccharide(LPS).Methods:The model was reproduced by LPS stimulating macrophages in culture in vitro.Induced NOS expression of macrophages was assayed by immuocytochemistry method,NO contents in medium were detected by Griess method,and the TNF-αcontents in medium were measured by indirect MTT method.Resutls:Induced NOS expression of macrophages,NO and TNF-αcontents in medium of macrophages were significantly increased after the stimulation by LPS.Herba Girardinia Ethyl acetate extract can remarkable ameliorate the inflammation factors increase.Conclusion:Herba Girardinia ethyl acetate extract inhibited TNF-αand NO excretions,and iNOS expression of macrophage,and then the present results provided the experimental base further development and research.
