【摘要】 目的:观察中药抑肺饮对肺腺癌细胞A549裸鼠移植瘤的抑制作用,探讨其治疗肺癌的作用机制。方法:将肺腺癌细胞A549接种于裸鼠腋窝皮下,形成移植瘤,待成瘤后分为阴性对照组(胃饲生理盐水)、阳性对照组(胃饲鸦胆子油乳)、抑肺饮高、中、低剂量组共5组,灌胃,27d后处死裸鼠,取下移植瘤,进行相关检测。采用流式细胞术检测细胞周期、CD95(Fas)、Bcl-2及凋亡率,免疫组织化学法检测Caspase-3、p16、VEGF、p53,TUNEL法检测凋亡指数。结果:中药抑肺饮中剂量组、阳性对照组的抑瘤率分别为:29.9%、35.9%,G0/G1期细胞百分比(65.7%±20.7%、63.6%±27.3%)均升高,p53(4.55%±1.48%、4.67%±0.79%)、p16(45.6%±19.5%、51.4%±29.1%)的表达阳性率均上升,VEGF(20.90%±8.44%、21.30%±9.28%)和Bcl-2(2.51%±1.37%、2.34%±1.68%)的表达阳性率均降低,凋亡率(27.7%±8.6%、30.5%±10.3%)和凋亡指数(1.53%±1.14%、1.47%±1.29%)均升高,CD95(Fas)(65.3%±9.77%、70.4%±14.2%)及Caspase-3(10.00%±4.26%、9.61%±3.66%)的表达率均上调,与阴性对照组比较,差异有统计学意义(P<0.05),中剂量组与阳性对照组各结果均无显著性差异(P>0.05)。结论:中药抑肺饮对肺腺癌细胞A549裸鼠移植瘤具有抑制作用,其机制可能与其抑制细胞增殖及诱导细胞凋亡有关。更多还原

【Abstract】 Objective:To observe the depressant effect of Chinese formula Yifei Yin on transplantation tumor of human lung adenocarcinoma A549 cells in nude mice,investigating the possible mechanism of the therapeutical effect of Yifei Yin on lung cancer.Methods:We inoculated the lung adenocarcinoma A549 cells on the nude mice to form transplantation tumor.The experimental nude mice with transplantation tumor were divided into five groups:negative control group,positive control group(oleum fructus bruceae group),high,moderate and low dose groups of Yifei Yin.Mice were intragastric administration and killed after 27 days.Transplantation tumor were taken and then detected.The cell cycle,CD95(Fas),Bcl-2 were detected by flow cytometry and Caspase-3,P16,VEGF,P53 were detected by immunohistochemical method,and the apoptotic index was detected by TUNEL method.Results:Moderate dose groups of Yifei Yin and positive control group can restrain the growth of the transplantation tumor obviously(tumor inhibitory rate 29.9%,35.9%),and increase the ratio of G0/G1 period(65.7%±20.7%,63.6%±27.3%).The expression of P53(4.55%±1.48%,4.67%±0.790%),P16(45.6%±19.5,51.4±29.1%),CD95(Fas),Caspase-3 of moderate dose groups of Yifei Yin and positive control group were up-regulated,and the expression of VEGF(20.9%±8.44%,21.3%±9.28%)and Bcl-2(2.51%±1.37%,2.34%±1.68%)of were both down-regulated.Apoptotic ratio(27.7%±8.60%,30.5%± 10.3%)and apoptotic index(1.53%±1.14%,1.47%±1.29%)of the two groups increased.The expression of CD95(Fas)(65.3%± 9.77%,70.4%±14.2%)and Caspase-3(10.0%±4.26%,9.61%±3.66%)of the two groups were both up-regulated also.Compared with negative control,the difference was all significant(P<0.05).The effect of the moderate dose group of Yifei Yin and positive control group were similar(P>0.05).Conclusion:Yifei Yin can restrain the transplantation tumor of human lung adenocarcinoma A549 cells in nude mice,and the mechanism may be related to inhibiting cell multiplication and inducing cell apoptosis.更多还原
