基质金属蛋白酶-2在高血压大鼠模型血管重塑中的表达及替米沙坦的作用

【摘要】 目的:探讨基质金属蛋白酶-2(MMP-2)在双肾一夹型高血压大鼠血管重塑中的表达及其调节机制。方法:将雄性Wistar大鼠随机分成对照组(假手术组)、双肾一夹型模型组、替米沙坦(Telmisartan)组3个组,其中Telm-isartan组每天给予药物灌胃。3组动物均在术后每周监测收缩压,8周后处死大鼠取胸主动脉与颈总动脉作形态学观察,免疫组化法检测颈总动脉与胸主动脉MMP-2的表达。结果:与对照组相比,模型组血压明显升高,血管中膜增厚,MMP-2表达显著增加;治疗8周后,Telmisartan组血压明显降低,管壁变薄,MMP-2表达减少。结论:MMP-2可能与高血压血管重塑有关,Telmisartan通过降低血压抑制血管中MMP-2的表达,并改善血管重塑。

【Abstract】 Objective:To explore the expression and the mechanism of MMP-2 in vascular remodeling of two-kidney and one-cliped(2KIC) hypertensive rats.Methods: Male Wistar rats were randomly divided into sham operation group(n=10),2KIC model group(n=10) and telmisartan group(n=7).Telmisartan group was treated with telmisartan by intragastric method every day.The blood pressure was measured in three groups every week.The morphologic changes in carotid artery and thoracic aorta were observed with staining HE after 8 weeks.The expression of MMP-2 in carotid artery and thoracic aorta was evaluated with immunohistochemisty.Results: Compared with those in the control group,the blood pressure,thickness of vessel wall and the expression of MMP-2 in the model group significantly increased.After treatment of Telmisartan,the blood pressure,thickness of vessel wall and the expression of MMP-2 in Telmisartan group significantly decreased.Conclusion: MMP-2 is likely to be related to vascular remodeling,and Telmisartan could inhibit the expression of MMP-2 by lowering the blood pressure and improve vascular remodeling in 2KIC hypertensive rats.
