成骨细胞承受不同大小和不同时间压力后增殖变化的研究
Osteoblast’s proliferation activity under different magnitudes and duration of mechanical stress in vitro
【摘要】 目的探讨不同大小和时间的压应力对成骨细胞的成活以及细胞周期的影响。方法将培养的MC3T3-E1成骨细胞分为加力组和对照组,使用离心机分别给加力组的细胞施加不同大小和不同作用时间的机械压力。用胎盘蓝细胞记数法检测成骨细胞的成活率,并且在成骨细胞受力后0h、24h和4d观察细胞数量的改变,同时使用流式细胞技术检测受力后成骨细胞细胞周期的改变。结果成骨细胞受到100g与200g压力后的增殖速度均大于对照组;持续30min和60min加压后的增殖速度也大于对照组;随着所受压力的增大和时间的延长,成骨细胞S期细胞百分比越高。结论 100g和200g压力以及持续30min和60min的压力能明显增加成骨细胞的增殖,同时细胞周期与细胞加力也有一定的关系。
【Abstract】 Objective To investigate the effect of different magnitudes and different time of mechanical stress on proliferation activity of mouse osteoblast-like cells(MC3T3-E1).Methods Compressive force by centrifugation was used to inflict MC3T3-E1 cells as the top-bottom axial stess,and 100g,200g,400g pressure were laid on MC3T3-E1 cells for 30,60 and 120 minutes,respectively.Cell proliferation was evaluated by cell counting.Cell cycle was measured by flow cytometry.Results There were more living cells in group of 100g and 200g and in group of 30 and 60 minutes than in the control.The S stage cells increased after compression.Conclusion The different magnitudes and different time of mechanical stress can influnce the proliferation and cell cycle in MC3T3-E1 cells.
