【摘要】 以铁皮石斛成熟种子为外植体,研究原球茎的诱导、增殖、分化和生根的最佳培养条件。结果表明:1/2 MS基础培养基最适合铁皮石斛的种子培养,45 d时原球茎诱导率达到95%;原球茎增殖的最佳培养基为1/2 MS+6-BA 0.5 mg/L+NAA 0.5 mg/L+20%马铃薯提取液,每代增殖16倍,原球茎形态好、变异少;原球茎团于1/2 MS+6-BA 2 mg/L+NAA 0.25 mg/L+20%马铃薯提取液的培养基上可诱导丛芽分化,于MS+NAA 0.5 mg/L+20%香蕉汁+活性炭5g/L的培养基上易于诱导不定根的形成,60 d后可形成健壮的完整小植株。更多还原

【Abstract】 The culture condition of protocorm induction,multiplication,differentiation and rooting from seeds of Dendrobium candidum was studied.The result showed that 1/2 MS with free plant growth regulator was the most optimum for seeds culture and the PLB percentage was 95%;1/2 MS containing 0.5 mg/L 6-BA,0.5 mg/L NAA and 20% tomato extract was best for PLBs’ multiplication,with 16 times of explant.PLBs can differentiate in 1/2 MS medium containing 2 mg/L 6-BA,0.25 mg/L NAA and 20% tomato extract.Buds can form plantlets after cultured for 60 days in MS medium containing 0.5 mg/L NAA,20% banana juice and 5 g/L active carbon.更多还原
