三白草总黄酮的超声波提取工艺研究

【摘要】 [目的]确定超声波提取三白草中总黄酮的最佳工艺条件。[方法]以三白草为原料,采用超声波技术提取其中的总黄酮,通过单因素试验研究乙醇浓度、料液比、超声温度、超声时间对总黄酮提取率的影响,通过正交试验对总黄酮的提取工艺进行优化。[结果]单因素试验结果表明,当乙醇浓度为60%,超声温度为55℃,超声时间为30min,料液比为1∶30时,黄酮提取率最高;正交试验结果表明,各因素对总黄酮提取率的影响依次为:超声温度>料液比>超声时间>乙醇浓度,总黄酮的最佳提取工艺为:提取溶剂70%乙醇,料液比1∶25,超声温度65℃、超声时间30min,此条件下总黄酮的提取量为6.890mg/g。[结论]该研究优化了超声波提取三白草中总黄酮的工艺参数。

【Abstract】 [Objective] The study was to determine the optimum extraction process of total flavonoids from Saururus chinensis(Lour)Bail with ultrasonic.[Method] With S.chinensis as the materials,the total flavonoids were extracted with ultrasonic technology.The effects of ethanol concentration,solid-liquid ratio,ultrasonic temperature and ultrasonic time on extraction rate of total flavonoids were studied through single factor test,and the extraction process was optimized through orthogonal test.[Result] The results of single factor test showed that when the ethanol concentration was 60%,ultrasonic temperature was 55 ℃,ultrasonic time was 30 min,solid-liquid ratio was 1∶30,the extraction rate of total flavonoids was the highest.The results of orthogonal test showed that the effects of each factor on extraction rate of total flavonoids in order was ultrasonic temperature>solid-liquid ratio>ultrasonic time>ethanol concentration.The optimum extraction process of total flavonoids was solid-liquid ratio of 1∶25,ultrasonic temperature of 65 ℃,ultrasonic time of 30 min with 70% ethanol as the extraction solvent,under this condition the extraction amount of total flavonoids was 6.890 mg/g.[Conclusion] The study optimized the extraction process of total flavonoids from S.chinensis with ultrasonic.
