IPPO催化氧化EGCG的催化特性研究

【摘要】 [目的]研究IPPO催化氧化EGCG的催化特性。[方法]以表没食子儿茶素没食子酸酯(EGCG)为原料,对固定化多酚氧化酶催化氧化形成儿茶素低聚物进行了探讨,并且研究以EGCG作底物时固定化多酚氧化酶的催化特性。[结果]固定化多酚氧化酶酶促氧化EGCG能够生成一种非茶黄素类的聚合物,此聚合物在pH值6.5、温度50℃、底物浓度1.5%、反应时间10min时生成量最大。[结论]在此条件下,其反应动力学符合米式方程,测得Vmax值为0.108OD/min,Km值为2.565mg/ml。

【Abstract】 [Objective]The study aimed to research the Characterization of Immobilized Polypehnol Oxidasecatalytic oxidation Epigallocatechin.[Method]With EGCG as material,oxidation of epigallocatechin which is the main component in catechins using immoblized poliphenol oxidase to find some unfound low-polymers and the characterization of immoblized poliphenol oxidase taking epigallocatechin as substrate were studied.[Result]The results showed that a sort of undefined substance which did not belong to theaflavains created in the process of oxidation of epigallocatechin using immoblized poliphenol oxidase. And the optimal conditions of producting this substance were pH 6.5,temperature 50 ℃,concentration of substract 1.5%,reaction time 10min.[Conclusion]The reaction kinetics accord with Michaelis-Menton model under the above conditions. The value of Vmax was 0.108 OD/min and Km was 2.565 mg/ml.
