两系杂交水稻纯度检测方法探讨

【摘要】 [目的]探索两系杂交水稻种子纯度的最佳检测方法。[方法]以丰两优香一号、丰两优四号和丰两优一号两系杂交水稻种子为材料,分别采用酯酶同工酶法和SSR分子法检测其纯度,并对2种方法的检测结果进行比较。[结果]同工酶测纯结果与SSR测纯结果的相关性较显著(r=0.964),但同工酶测纯结果总体低于SSR测纯结果;了解种子的大致纯度时可采用同工酶方法测纯,精确掌握种子纯度时可采用SSR分子方法测纯。[结论]SSR测纯结果更接近种子的真实纯度。

【Abstract】 [Objective] The aim was to explore the optimum detection method on seed purity of two-line hybrid rice.[Method] With the seeds of two-line hybrid rice varieties Fengliangyouxiang1,Fengliangyou4 and Fengliangyou1 as the materials,their purities were determined with the methods of esterase isozyme and SSR,and the detection results of the 2 methods was compared.[Result] The correlation of the detection results of the 2 method was significant,but the detection result of esterase isozyme method was lower than that of SSR method.The esterase isozyme method could be used to determine the approximately purity of the seeds,and SSR method could be used to accuratly determine the purity of the seeds.[Conclusion] The detection result of SSR method was more close to the real purity of the seeds.
