分光光度法测定酯和内酯含量的研究
Determination of Ester and Lactone Based on Spectrophotometry
【摘要】 [目的]研究酯和内酯的分光光度检测方法。[方法]首先,酯和内酯在碱性条件下与羟胺反应生成异羟肟酸;然后,在酸性条件下异羟肟酸又与Fe3+络合生成紫红色的异羟肟酸铁络合物;最后,在506nm波长下间接测定酯和内酯含量。[结果]相同浓度的酯或内酯,与其相应的络合物吸光度相同,且络合物吸光度与其相应的酯或内酯的浓度呈良好的线性关系,标准曲线回归方程为y=4.1186x+0.0067(R2=0.9996),平均回收率为102.5%,相对标准偏差RSD为0.64%。[结论]分光光度法具有较高的灵敏度、准确度和精密度,适用于各种酯或内酯及混合酯总量的测定。
【Abstract】 [Objective] The aim of this research was to study the detection method of ester and lactone by spectrophotometry.[Method] First of all,esters and lactone reacted with hydroxylamine and formed hydroxamic acid under alkaline conditions;secondly,hydroxamic acid reacted with Fe3+ and generated amaranthine hydroxamic acid-iron complex;finally,esters and lactone contents were measured indirectly at the wavelength of 506 nm.[Result] The results were found that the absorbance of complex was invariable in the same concentration of ester or lactone and had a good linear relationship.The standard curve regression equation was y=4.118 6 x+0.006 7(R2=0.999 6) and the average recovery and relative standard deviation (RSD) were 102.5% and 0.64%,respectively.[Conclusion] The method has higher sensitivity,accuracy and precision,could be applied to determine the contents of carboxylic ester,lactone and their complexes.
