组织因子途径抑制物-2表达与肝癌细胞体外侵袭能力的实验研究

【摘要】 目的:研究肝癌组织中组织因子途径抑制物-2(tissue factor pathway inhibitor-2,TFPI-2)基因的表达,同时探讨TFPI-2对肝癌细胞侵袭能力的影响。方法:应用原位杂交法和免疫组织化学法检测30例肝癌患者的癌组织和癌旁正常肝组织中TFPI-2基因和蛋白的表达;通过脂质体法将TFPI-2基因转染人HepG2细胞,RT-PCR检测转染前后TFPI-2 mRNA的表达;Transwell小室法测定TFPI-2基因转染前后肝癌细胞体外侵袭迁移能力的变化。结果:肝癌组织中TFPI-2mRNA与蛋白的表达水平明显低于癌旁正常肝组织(P<0.05);成功转染TFPI-2基因的肝癌细胞HepG2中证实有TFPI-2 mRNA的表达;与未转染的细胞相比,转染TFPI-2的细胞体外侵袭能力明显下降,穿膜细胞数分别为45.2±5.6和87.8±4.5,差异有统计学意义(P<0.05);而迁移能力无明显变化,转染前后HepG2细胞的穿膜细胞数分别为140.4±7.4和152.8±9.6,差异无统计学意义(P>0.05)。结论:肝癌组织中TFPI-2表达明显降低,TFPI-2基因表达可明显抑制肝癌细胞的体外侵袭能力。

【Abstract】 Objective:To study the expression of tissue factor pathway inhibitor-2(TFPI-2) in hepatocellular carcinoma (HCC) and explore the effect of TFPI-2 on cell invasion and migration of HCC. Methods:The expressions of TFPI-2 mRNA and protein were determined by in situ hybridization (ISH) and immunohistochemical staining (IHC),respectively,in 30 cases of HCC and 30 cases of tumor-adjacent normal tissues. The expression vector pcDNA3.1-TFPI-2 was transfected into human HCC cancer cell line,HepG2 cells,via liposome mediation. TFPI-2 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The invasive and migrative behaviors of transfected and nontransfected cells were assessed using Transwell chamber method. Results:The expression levels of TFPI-2 mRNA and protein were obviously lower in HCC tissues than that in tumor-adjacent normal tissues (P<0.05). Transcription of TFPI-2 mRNA was validated in transfected cells. The invasion capability of TFPI-2-positive cells was obviously decreased compared with that of TFPI-2-negative cells (45.2±5.6 vs 87.8±4.5,P<0.05); but the migration capability had no significant difference before and after transfection (140.4±7.4 vs 152.8±9.6,P>0.05). Conclusion:The expression of TFPI-2 gene is obviously lower in HCC tissues than that in normal hepatic tissues. Expression of TFPI-2 gene greatly can inhibit the invasive ability of HCC cells in vitro.
