日本粗榧果肉中三种二萜类化合物体外抗癌活性研究
Effects of 3 Diterpenes from Torreya nucifera on Inhibiting Proliferation of Cultured Human Leukemia K562 Cells in vitro
【摘要】 目的:观察从日本粗榧果肉中分离得到的三种二萜类化合物18-hydroxyferruginol(A)、kayadiol(B)及hinokiol(C)对人白血病细胞K562和鼠肉瘤细胞S180生长的影响。方法:以人白血病细胞K562为研究对象,采用MTT法检测上述三种二萜类化合物对K562细胞生长的抑制率,用相差倒置显微镜观察细胞形态变化。根据检测结果,对三种化合物进行抗癌活性筛选,将筛选出的有效化合物kayadiol再作用于鼠肉瘤细胞S180,观察其对S180细胞的作用。结果:18-hydroxyferruginol及kayadiol对K562细胞有抑制其增殖的作用;其有效浓度分别是1×10-4mol/L和1×10-8mol/L~1×10-4mol/L,kayadiol对S180细胞也有一定的抑制作用。hinokiol对于K562细胞的增殖没有明显的抑制作用。结论:日本粗榧果肉中分离得到的二萜类化合物18-hydroxyferruginol(A)、kayadiol(B)对人白血病细胞K562的生长具有一定的抑制作用。
【Abstract】 Objective:To study the effects of 3 diterpenes (18-hydroxyferruginol (A)、kayadiol(B) and hinokiol(C) ) from Torreya nucifera on Inhibiting proliferation of cultured human Leukemia K562 cells in vitro . Methods :MTT was used to observe the effect of inhibiting proliferation of human Leukemia K562 cultured in vitro with the 3 diterpenes of different concentration in it for different periods of time . Results :It was found that compounds A had the effects on inhibiting proliferation of human Leukemia K562 cultured for 12h, 24h and 36h at dose of 1×10-4mol/L ,and for compound B ,cultured for 24h, 1×10-4mol/L~1×10-4mol/L were effective; for 48h 1×10-4mol/L~1×10-8ol/L were effective ,and for 72h 1×10-6ol/L~1×10-7ol/L were effective. Compound C had no effect. In addition , compound B had the effect on inhibiting proliferation of S180 cells cultured for 72h at dose of 1×10-6mol/L. Conclusion : Diterpenes 18-hydroxyferruginol (A) and kayadiol(B) have the effects on inhibiting the proliferation of cultured human Leukemia K562 cells in vitro. kayadiol(B) is more effective.
