天麻醒脑胶囊对血管性痴呆动物认知功能障碍的影响

【摘要】 目的:研究天麻醒脑胶囊对大鼠血管性痴呆所致的认知功能障碍的影响及其初步机理。方法:四血管阻断法复制大鼠脑缺血痴呆模型,采用跳台、避暗和水迷宫方法分别评价天麻醒脑胶囊对血管性痴呆所致的认知功能障碍的影响,运用试剂盒测定脑组织丙二醛(MDA)、乙酰胆碱酯酶(AChE)、脂褐质的水平。结果:跳台和避暗方法测试结果表明,天麻醒脑胶囊(0.6、1.2g/kg)明显减少5min内错误次数明显,显著延长错误潜伏期;水迷宫方法测试表明,0.6和1.2g/kg的天麻醒脑胶囊明显减少大鼠进入盲端的错误次数,显著缩短游完全程达岸时间。与模型组比较,2.4g/kg的天麻醒脑胶囊显著降低脑组织AChE活性、MDA及脂褐质含量。结论:天麻醒脑胶囊通过降低血管性痴呆大鼠脑组织Ach水平、减少自由基的产生而显著改善血管性痴呆所致大鼠学习记忆功能障碍。

【Abstract】 Objective:To investigate the effects of Tianma xingnao capsule on obstacle of learning and memory function induced by vascular dementia.Methods:The vascular dementia(VD)model of rats was prepared by permanent occlusion of four-lateral common carotid arteries.The methods of avoidance response to darkness,springing to platform after electric stimulation and water maze were used to investigate the effects of Tianma xingnao capsule on the dementia model,respectively.The activity of acetylcholine esterase(AChE)and the contents of malondiadehyde(MDA)and lipofuscin in brain tissue were measured by biochemical methods and fluorospectrophotometry,respectively.Results:In the tests of avoidance response to darkness and springing to platform after electric stimulation,Tianma xingnao capsule at the doses of 0.6 and 1.2 g/kg could significantly decrease the error times within 5 min in dysmnesia groups and prolong the incubation periods of memory.Compared with the model group,the mice swimming time and the error times within 3 min in the test of water maze were markedly shorter in the groups of high and middle doses of Tianma xingnao capsule.Tianma xingnao capsule,as compared with the model group,decreased the level of MDA and the activity of AChE,and obviously inhibited the increase of the lipofuscin in brain tissue.Conclussion:Tianma xingnao capsule could improve the abilities of learning and memory in animals with vascular dementia.The preliminary mechanisms may be associated with its effects on decreasing the activity of AChE,promoting the function of cerebral monoamine nervous system in brain tissue and reducing the level of lipid peroxidation in brain tissue.
