黄芩苷对糖尿病肾病大鼠血浆及肾组织AngⅡ的影响
Effect of Baicalin on Plasma and Renal AngⅡ in Diabetic Nephropathy Rats
【摘要】 目的研究黄芩苷对糖尿病肾病(DN)大鼠AngⅡ的影响,并从AngⅡ的非血流动力学机制探讨黄芩苷防治DN的作用。方法取SPF级雄性SD大鼠19只,随机分为3组,正常组5只,模型组7只,黄芩苷组7只。模型组和黄芩苷组大鼠单次大剂量腹腔注射链脲佐菌素(STZ)7周后造成DN大鼠模型。成模后,黄芩苷组大鼠每天腹腔注射黄芩苷水溶液40mg/kg;模型组大鼠每天腹腔注射注射用水1mL/只;正常组大鼠不进行腹腔注射。治疗6周后,所有大鼠眼眶取血,用放射免疫法检测血浆AngⅡ的含量;取一侧肾置于4%多聚甲醛固定液中,采用免疫组化法检测肾小管TGF-β含量;另一侧肾取肾皮质匀浆后,-20℃保存,用放射免疫法检测肾组织AngⅡ的含量。结果黄芩苷组TGF-β的表达数量减少、强度减弱,与模型组比较,具有显著性差异(P<0.05),与正常组比较,差异无统计学意义。黄芩苷组血浆AngⅡ降低,与模型组相比较,具有显著性差异(P<0.05),与正常组比较,差异无统计学意义。肾组织AngⅡ与模型组比较及与正常组比较,差异均无统计学意义。结论黄芩苷具有保护肾脏的作用,其作用机制可能是通过降低血浆AngⅡ的含量来抑制肾小管TGF-β的表达,从而延缓DN的发展。
【Abstract】 Objective To study effects of baicalin(Bai) on AngⅡ in diabetic nephropathy(DN) rats and to explore its preventive and therapeutic effect on treating DN from the non-hemodynamic mechanism of AngⅡ.Methods Nineteen specific pathogen-free male rats were randomly divided into three groups:normal control group(5 rats),diabetic nephropathy model control group(7 rats),baicalin treated group(7 rats).DN rat model was established by one-dose intraperitoneal injection of streptozotocin.Seven weeks after the modeling,rats in baicalin group recieved intraperitoneal injection of baicalin solution(40 mg/kg),rats in DN model group received intraperitoneal injection of normal saline(1 mL for each rats),and the normal control group did not receive any intraperitoneal injection.After treatment for 6 weeks,orbital blood from all rats was collected to determine plasma AngⅡ by radioimmunoassay.Unilateral kidney was processed by 4 % paraformaldehyde fixation and then was used for the determination of TGF-β by immunohistochemical assay.The other unilateral kidney was got out to prepare for renal cortex homogenate,and then was used for the determination of renal tissue AngⅡ by radioimmuno assay.Results The quantity and degree of TGF-β expression in baicalin group was decreased,and there was statistical significance between baicalin group and the model group(P < 0.05),but the difference has no significant between baicalin group and normal group.Plasma AngⅡ in baicalin group was significantly decreased,and the difference was significant compared with the model group(P < 0.05),while was insignificant compared with the normal group.There was no significant difference of renal AngⅡ among the three groups.Conclusion Decrease of plasma AngⅡ and inhibition of renal TGF-β expression may be the renal protective mechanisms of baicalin,which delays the progress of DN.
