石榴皮提取物对小鼠实验性前列腺增生的影响
Effects of Pericarpium Granati Extracts on Mice Benign Prostatic Hyperplasia
【摘要】 目的研究石榴皮提取物(Pomegranate peel extracts,PPE)对丙酸睾酮诱发小鼠实验性前列腺增生(Benign prostatic hyperplasia,BPH)的影响。方法采用皮下注射丙酸睾酮诱发小鼠实验性前列腺增生模型,以250及500mg/kg两种不同剂量的石榴皮提取物灌胃给药,连续12d,测定小鼠前列腺湿重及前列腺指数,光镜下观察前列腺组织形态学变化,并测定血清睾酮(T)水平,前列腺组织中一氧化氮(NO)含量、丙二醛(MDA)含量、抗氧化能力指数(ORAC)水平,HPLC法测定石榴皮提取物体外对5α-还原酶活性的影响。结果与模型组相比,石榴皮提取物可显著降低前列腺指数并改善其病理状态,同时降低血清T水平、血清NO含量和前列腺组织中MDA含量,提高抗氧化能力指数(P<0.05或P<0.01)。此外,体外实验证明石榴皮提取物对5α-还原酶活性具有一定的抑制效果。结论石榴皮提取物对丙酸睾酮引起的小鼠实验性前列腺增生具有一定的抑制作用,其作用机制可能与抑制5α-还原酶活性及调节机体氧化应激状态有关。
【Abstract】 Objective To study the effects of pomegranate pericarp extracts(PPE)on experimental benign prostatic hyperplasia(BPH) in mice.Methods The BPH model was established by subcutaneous injection of testosterone propionate in mice.After treatment for 12 continuous days,the mice were sacrificed to determine the index of prostate gland(PI) and to observe the morphological changes in the prostate.The level of testosterone(T)in plasma,the content of NO,Malondialdehyde(MDA)and oxygen radical absorbance capacity(ORAC) in the prostate were also determined.Meanwhile,the inhibition of 5α-reductase was observed in vitro.Results Compared to the BPH model group,the prostatic wet weight and PI were decreased after PPE treatment for 12 days.Meanwhile,ORAC level in the prostate was increased,and prostatic MDA level and plasma NO and T levels were decreased(P < 0.05 or P < 0.01).In addition,PPE could also inhibit the activity of 5α-reductase in vitro.Conclusion PPE have protective effect on mice BPH induced by testosterone propionate.The mechanisms are probably related to the inhibition of 5α-reductase and regulation of oxidative stress.
