痰热清注射液对脓毒症大鼠炎症介质及生存率的影响

【摘要】 目的观察痰热清注射液对脓毒症大鼠高迁移率族蛋白B1(HMGB1)水平及生存率的影响。方法采用盲肠结扎穿孔术(CLP)制备脓毒症模型。112只Wistar大鼠,其中16只随机分为模型组及痰热清治疗组,并观察大鼠术后7d生存率,另96只大鼠按随机数字表法分为正常组、假手术组、模型组和痰热清治疗组。用ELISA法检测各组血浆HMGB1和IL-6的水平。结果痰热清治疗组7d生存率明显高于模型组。与假手术组比较,模型组大鼠血浆HMGB1在CLP后12h明显升高,48h达到高峰,12、24及48h3个时点与假手术组比较差异显著性,痰热清治疗组在上述3个时点可明显降低HMGB1的水平;IL-6水平在CLP后6h明显升高,12h达到高峰,除2h时点外,各时点与假手术组比较差异显著;与模型组比较,除2h时点外,痰热清治疗组IL-6水平明显降低。结论痰热清注射液可明显抑制脓毒症大鼠血浆HMGB1及IL-6水平,提高脓毒症大鼠的生存率。

【Abstract】 Objective:To explore the effects of Tanreqing Injection on the level of high mobility group protein B1(HMGB1) and survival rate of rats with sepsis.Methods:The sepsis model was prepared by CLP method.112 Wistar rats were selected.16 rats were randomly divided into the model group and the Tanreqing Injection treatment group for the observation of survival rates after postoperative 7d.Another 96 rats were randomly divided into 4 groups as follows: sham operation group,normal control group,model group and Tanreqing Injection treatment group.The levels of HMGB1 and IL-6 in every group were examined by ELISA method.Results:Compared with the model group,the survival rates of rats in Tanreqing Injection treatment group at 7d were notably raised.Compared with the sham operation group,the levels of HMGB1 in the model group increased significantly at 12h,which peaked at 48h after CLP.At the time of 12h,24h and 48h after CLP,the levels of HMGB1 in the Tanreqing Injection treatment group decreased significantly compared with the model group.Compared with the sham operation group,the level of IL-6 in the model group increased significantly except the time point of 2h,which peaked at 12h.Compared with the model group,the level of IL-6 in the Tanreqing Injection treatment group decreased significantly except the time point of 2h.Conclusion:Tanreqing Injection can obviously decrease the level of HMGB1 and IL-6 and raise the survival rate of rats with sepsis.
