银杏叶提取物预处理对小鼠局灶性脑梗死的保护作用
Experimental Study on the Protective Effect of Ginkgo Biloba Extract in Mice after Local Cerebral Infarction
【摘要】 目的观察银杏叶提取物(GBE)对小鼠局灶性脑梗死是否具有保护作用。方法健康成年雄性昆明小鼠随机分为假手术组、模型组以及GBE预处理组。用线栓法制作小鼠永久性大脑中动脉闭塞模型。假手术组及模型组术前30min腹腔注射0.9%氯化钠注射液0.4ml,GBE预处理组腹腔注射舒血宁35mg/kg。按八级行为评分法记录小鼠清醒后神经功能缺损程度,TTC染色测定梗死体积,黄嘌呤氧化酶法测定SOD1活性,TBA法测定丙二醛(MDA)含量,TUNEL测定细胞凋亡。结果与模型组相比,GBE预处理组小鼠术后24h神经功能缺损评分下降,梗死体积缩小,SOD1活性升高,MDA含量减少,神经元凋亡减轻。结论GBE对小鼠局灶性脑梗死具有一定保护作用,提高SOD1活性为其发挥保护作用的机制之一。
【Abstract】 Objective:To explore the protective effect of ginkgo biloba extract(GBE) in mouse after local cerebral infarction.Methods:Healthy adult male Kunming mice were randomly divided into three groups: Sham-operated group,model group and GBE pro-condition group.The model of permanent middle cerebral artery occlusion(PMCAO) was set up.30min before operation,0.4ml 0.9% sodium chloride and GBE of 35 mg/kg were intraperitoneally injected into the mice in each group respectively.After the consciousness recovery,the animals were scored using an eight-grade neurological function scale,the sections were stained by TTC for observing the infarct volume,the leves of SOD1 and MDA were measured,the used of TUNEL stained to inverstigate the apoptotic cells.Results: Compared with model group,the score of neurological function in GBE pro-condition group was lower,the infarct size smaller,the level of SOD1 higer,MDA decreased,the apoptotic released.Conclusion: GBE can protect murine local cerebral infarction in some degree,elevating the activity of SOD1 is one of the mechanism.
