半夏超临界CO2乙醇萃取物对皮层定位注射青霉素诱发大鼠痫性放电和海马区癫痫相关递质的影响
The Effects of Supercritical-CO2 Alcohol Fluid Extractions of Pinellia Tuber on the Epileptiform Discharge and Related Neurotransmitters in Hippocampus in Seizure Rats Induced by Penicillin Localized Injected in Cortex
【摘要】 目的:研究半夏超临界CO2乙醇萃取物对大鼠皮层定位注射青霉素诱发惊厥模型的对抗作用,并探讨其作用机制。方法:建立大鼠皮层定位注射青霉素惊厥模型,用RM6240C型多道生理信号采集处理系统记录脑电图,以托吡酯(Topiramate,TPM)为阳性对照,观察灌胃(ig)给药对惊厥行为和脑电图的影响;采用HPLC法测定海马区Glu、Asp、Gly、GABA的含量变化。结果:半夏超临界CO2乙醇萃取物(29.96g.kg-1ig)可延长痫性发作潜伏期、减轻发作程度,延长痫性放电的潜伏期,减少痫波发放频率,减小放电最高波幅,分别与模型组比较,均具有统计学差异(P<0.05或<0.01)。此外,该药能使大鼠海马区Glu含量降低,GABA含量显著升高,与模型组比较均具有统计学差异(P<0.05或<0.01),而Asp、Gly含量与模型对照组间均无统计学意义(P>0.05)。结论:半夏超临界CO2乙醇萃取物能对抗大鼠皮层定位注射青霉素诱发痫性放电,其抗惊厥作用性质和TPM相似。作用机制与增加大鼠海马GABA的含量,提高GABA功能,并能降低Glu的含量,限制Glu兴奋功能有关。
【Abstract】 Objective:To investigate the antagonism of supercritical-CO2 alcohol fluid extractions of Pinellia Tuber on the mode of seizure rats induced by penicillin and explore its mechanism of anticonvulsant action.Methods:Using the model of seizure rats induced by penicillin localized injected in cortex investigated the effect of SFE-CO2 alcohol fluid extractions of Pinellia Tuber on the convulsant behaviors and hippocampus EEG(Recoded by RM6240C multichannel physiological signal collection and analysis recorder).The neurotransmitters(Glu,Asp,Gly and GABA)of hippocampus were determined by high performance liquid chromatography(HPLC).Results:SFE-CO2 alcohol fluid extractions of Pinellia Tuber could prolong the latent period on the penicillin-induced seizure and lighten the extent of seizure.At the same time,it could prolong latent period of epileptiform discharge,reduce the frequency of epileptiform discharge,decrease the highest wave of hippocampus EEG..All above observing indexes compared with the model control groups were significantly difference(P<0.01).Moreover,it might have been increase the level of GABA and decrease Glu level(P<0.05 or P<0.01),but no statistics significant on the level of Gly,Asp(P>0.05).Conclusion:SFE-CO2 alcohol fluid extractions of Pinellia Tuber could antagonize penicillin-induced seizure in rats.Its anticonvulsant properties resembled TPM.The anticonvulsant mechanism of it might have been due to increase the level of GABA and decrease Glu level.
