姜黄素诱导人胃腺癌BGC-823细胞凋亡的光动力学研究

【摘要】 目的:研究光照时间和波长与光敏化姜黄素诱导细胞凋亡效应之间的关系。方法:体外培养的胃腺癌BGC-823细胞加姜黄素后光照0、4、8、12、24h,4、8、12h组部分样品光照结束立即流式细胞术检测凋亡率,其余样品继续避光处理至24h检测凋亡率;BGC-823细胞加姜黄素后进行全波长及400、470、530、660nm的波长光照处理,24h后流式细胞术检测凋亡率。结果:光照对姜黄素诱导BGC-823细胞凋亡的增效作用与光照时间有关。低浓度姜黄素避光处理24h不能诱发细胞凋亡,光照4、8、12h立即检测组的凋亡率显著低于光照结束后避光处理到24h组,而光照后继续避光处理组中,4、8h组效果显著低于12h以上组。姜黄素要获得显著的凋亡增敏效果需要400nm左右波长的光照,可见光全波长的对姜黄素诱导细胞凋亡效果最佳。结论:低浓度姜黄素诱导BGC-823细胞凋亡需要一定的光照时间及药物作用时间,12h以上的光照时间是引发最大凋亡必需的,光照波长越接近最大激发波长凋亡诱导作用越强。

【Abstract】 Objective:To investigate the relation of apoptosis effects induced by curcumin and the time and wavelength of light needed.Methods:BGC-823 cell was treated by curcumin in light respectively for 0 hour,4hour,8hours,12 hours and 24 hours.Flow cytometry was used to measure the apoptosis rate of BGC-823 cells.For the groups treated respectively for 4 hours,8 hours and 12 hours in light,one half were measured the apoptosis rate immediately after the ending of treatment in light,the other were put in dark after that till 24 hours.All groups left were measured the apoptosis rate of BGC-823 cells at 24 hours.BGC-823 cell was also treated by curcumin respectively in visible light or light with a series of wavelength of 400nm,470nm,530nm and 660nm for 24 hours and then was measured for the rate of apoptosis.Results: Apoptosis can not be induced at low concentration of 5μmol/L in dark till 24 hours.For the groups treated respectively for 4 hours,8 hours and 12 hours in light,more marked apoptosis effect of curcumin can be seen in groups with next dark treatment than without.After Long time treatment of 12 hours or 24 hours in light,more marked apoptosis effect can be seen compared with shorter time treatment of 4 hours or 8 hours when measuring the apoptosis rate of BGC-823 cells at 24 hours.We also found 400nm wavelength light or visible light can induce more marked apoptosis effect than light of other wavelength.Conclusion: We can infer unalterably that some time in light and enough time treated with curcumin are necessary for apoptosis induced at low concentration of 5μmol/L.It is needed that at least 12 hours and the light with wavelength closing to the excitation we can get the most marked effect of apoptosis.
