灵芝多糖对失血性休克再灌注肺损伤的保护作用
Protection of GLP on Lung Injury in Hemorrhagic Shock Reperfusion
【摘要】 目的:探讨灵芝多糖(GLP)对失血性休克再灌注(SH-R)复苏时肺损伤的保护作用及机制。方法:复制家兔失血性休克再灌注复苏模型,随机分成假手术组(S组)、生理盐水再灌注复苏组(NS组)和质量分数为1%的GLP再灌注复苏组(LS组),于放血前、休克40min、再灌注复苏40min和90min时观察动脉血氧分压(PaO2)、血浆、肺组织丙二醛(MDA)、肿瘤坏死因子α(TNF-α)含量、肺组织超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)活性、肺组织湿/干重比(W/D)、肺泡损伤率(IAR)、肺组织病理学变化及肺组织细胞的凋亡情况。结果:①休克40min时,PaO2明显降低,血清MDA和TNF-α则明显升高,当用NS再灌注时,PaO2进一步降低,血清MDA和TNF-α进—步升高,而当用LPS再灌注时,PaO2、MDA和TNF-α变化不明显,但与NS组同时间点相比差异显著(P<0.05);②NS组和LS组肺组织SOD活性明显低于S组,而LS组又明显高于NS组(P<0.05);NS组和LS组肺组织MPO活性、MDA、TNF-α含量、W/D、IAR、肺泡Ⅱ型上皮细胞凋亡百分数明显高于S组(P<0.05),而LS组又明显低于NS组(P<0.05);③病理显示:NS组肺损伤明显,LS组肺损伤不明显。结论:GLP对SH-R中肺损伤有保护作用。
【Abstract】 Objective:To study the protection of GLP on lung injury in hemorrhagic shock reperfusion(HS-R).Methods:With the model of HS-R in rabbits,the animals were divided into three groups:sham operation group(S group),reperfusion with NS group(NS group),reperfusion with 1% GLP group(LS group).The changes of PaO2,MDA,TNF-α,SOD,MPO,W/D,IAR in serum and lung were observed at the time of pre-shock,shock 40 min,post-reperfusion 40 min and 90 min respectively.The apoptosis of lung were examined too.Results:①PaO2 decreased significantly,but the content of MDA and TNF-α in serum increased signi ficantly at the time of shock 40 min(P<0.05).When reperfusion with NS,the above changes were more obvious;but when reperfusion with LPS,the changes were not obvious.②The activity of SOD in lung of NS group and LS group were significanly lower than that of S group(P<0.05),but its activity in LS group was significantly higher than that of NS group(P<0.05).The level of MDA,TNF-α,MPO,W/D,IAR and the apoptotic rate of alveolar epithelial cells in lung of NS group and LS group were significantly higher than that of S group(P<0.05),but their conents in LS group were significantly lower than that of NS group(P<0.05).③The pathological results showed that serious pulmorary structural injury occurred in NS group,but no significnt injury was found in LS group.Conclusion:GLP has protective effects on lung injury in HS-R.
