四逆汤含药血清对阿霉素诱导心肌细胞损伤机制影响的研究
Effect of Blood Serum of Decoction for Treating Yang Exhaustion on by Cardio-toxicity in Myocardial Cell Induced Adriamycinin
【摘要】 目的:观察四逆汤含药血清对阿霉素(ADR)引起的培养乳鼠心肌细胞毒性的影响。方法:采用原代培养的Wister大鼠乳鼠心肌细胞,以1.0μg/mL的ADR造成急性心肌细胞中毒模型,测定心肌细胞乳酸脱氢酶(LDH)漏出量、心肌细胞丙二醛(MDA)含量、心肌细胞搏动频率及MTT分析法测定心肌细胞活力,测定过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)、超氧化物歧化酶(Cu-SOD),观察大鼠生存率的变化。结果:四逆汤含药血清对ADR引起的心肌细胞损伤可减少LDH漏出量和心肌细胞内MDA含量,并改善心肌细胞活力和搏动频率,改善Cu-SOD的活性,延长大鼠生存期。结论:四逆汤含药血清对ADR引起的心肌细胞损伤有一定的保护作用,其作用机制与其抑制ADR引起的脂质过氧化损伤有关。
【Abstract】 Objective:To observe the effect of decoction for treating yang exhaustion on adriamycin in induced cardiotoxicity in cultured myocardial cells.Methods:Primary cultured myocardial cells of Wister neonate rats were treated with adriamycinin hydrochloride(1.0μg/mL)and decoction for treating yang exhaustion.The content of lactate dehydrogenase(LDH)leakage,malondialdehyde(MDA)in myocardium,myocardial cell activity and its beating rate were assayed.CAT,T-SOD,Cu-SOD of rat myocardium were detemined.Observe every groups survival rate.Results:Decoction for treating yang exhaustion could decrease significantly the contents of MDA and LDH induced by adriamycin and imp rovemyocardial cell activity and its beating rate.Conclusion:Decoction for treating yang exhaustion has protective effect on adriamycinin induced cardiotoxicity in cultured myocardial cells,which may be related to inhibiting lipid peroxidative damage.
