宁动颗粒治疗抽动秽语综合征的药效学研究

【摘要】 目的探讨宁动颗粒对抽动秽语综合征的药效学及其毒性的影响。方法采用宁动颗粒灌胃法观察对小鼠自主活动的影响、对阈下剂量戊巴比妥钠致小鼠睡眠率的影响、对去水吗啡(APO)引起小鼠攀爬行为的影响、对小鼠脑内多巴胺(DA)含量的影响及对小鼠急性毒性的影响。结果宁动颗粒高、低剂量均可明显抑制小鼠的自主活动能力;高剂量有协同阈下剂量戊巴比妥钠致小鼠睡眠的作用;高、低剂量均有明显抑制APO引起的小鼠黑质-纹状体系统内DA功能增强的作用,明显缩短持续垂直攀爬时间;高剂量可以降低脑内DA含量,低剂量也有降低脑内DA含量的作用趋势。急性毒性试验1d内给小鼠药量相当于儿童用量的77.3倍,小鼠生存良好。结论药理学实验证明宁动颗粒能降低脑内DA含量,有镇静、拮抗APO引起的攀爬行为增强及助睡眠作用;毒理学表明宁动颗粒无明显毒副反应。

【Abstract】 Objective To study the pharmacodynamics and toxicity of Ningdong granule for Tourette syndrome (TS). Methods Ningdong granule was given to the mouse by intragastric administration.After that, observe the effect on autonomic activities of mouse,the mouse sleep rate at subthreshold dose of sodium pentobarbital,the special climbing behavior caused by Apomorphine(APO), the dopamine(DA) content in mouse brain and the acute toxicity. Results Both the high and low dose of Ningdong granule could obviously inhibit mouse special climbing behavior.High dose could function together with subthreshold dose of sodium pentobarbital to cause mouse sleep.Both the high and low dose could inhibit DA function of mouse substantia nigra-corpus striatum,and could obviously shorten mouse continual vertical creeping.The high dose could lower brain DA content.The low dose had the tendency of lowering brain DA content.In the acute toxicity test,the dose corresponding to 77.3 times of children dose was given to the mouse.The mouse lived well. Conclusion Ningdong granule can lower brain DA content.It has the function of sedation,antagonizes creeping caused by APO and helps sleep.Toxicology certifies that it has no obvious toxic action.
