13-甲基肉豆蔻酸抗肿瘤作用的研究
Antitumor activity of 13-methyltetradecanoic acid
【摘要】 目的:研究13-甲基肉豆蔻酸(13-MTD)的抗肿瘤作用。方法:应用台盼蓝排染法、MTT法测定13-MTD对人乳腺癌Bcap-37等6种体外培养肿瘤细胞的细胞毒作用。观察13-MTD对小鼠移植性实体瘤S180的瘤重抑制率,对白血病P388荷瘤鼠的生命延长率。观察13-MTD对正常及免疫抑制小鼠免疫机能的影响。结果:6种肿瘤细胞株的IC50:13.9~33.0μg·mL-1。400mg·kg-1灌胃对小鼠肉瘤S180的瘤重抑制率为71.74%,对白血病P388荷瘤鼠的生命延长率为41.49%。对机体的免疫机能未见抑制作用,对环磷酰胺(CTX)诱发的免疫抑制,不仅不致恶化,个别指标如胸腺系数等还有缓解作用。结论:13-MTD有较强的体内外抗肿瘤活性。对机体免疫机能未见明显影响,对CTX诱发的免疫抑制状态,联用13-MTD未见恶化现象。
【Abstract】 Objective:To investigate the antitumor effect of 13-methyltetradecanoic acid(13-MTD).Methods:Both in vitro and in vivo tests for antitumor activity were conducted,and the effect of 13-MTD on immune functions was examined.Results:In in vitro test,the IC50 ranged from 13.9 to 33.0 μg·mL-1 for six tumor cell lines,including mammary adenocarcinoma Bcap-37 cells.In in vivo test,intragastric 13-MTD 400 mg·kg-1 per day inhibited tumor size by 71.74% in solid tumor model of S180,and extended life-span by 41.49% in leukemia model of P388.13-MTD had no significant effect on immune function of normal mice.When combined with cyclophosphamide(CTX),13-MTD did not aggravate the immune suppression induced by CTX,but actually reversed such immune suppression in some indexes such as thymus index.Conclusion:In both in vitro and in vivo tests,oral 13-MTD has significant antitumor activity for growth of cancer cell lines and xenografted tumors in mice.This agent does not suppress immune functions,nor aggravate the immune suppression induced by combination treatment with CTX.[
