羟萘酸沙美特罗合成新工艺研究
New synthesis process of salmeterol xinafoate
【摘要】 目的:合成抗哮喘药羟萘酸沙美特罗。方法:以水杨醛和4-苯基-1-丁醇为原料,经酰化、缩合、还原、催化氢化脱苄、成盐等反应合成羟萘酸沙美特罗。结果:所得产物结构经红外光谱、核磁共振谱及质谱等确证,以水杨醛计算总收率为31.5%。结论:该方法简化了操作步骤,优化了反应条件,降低了成本,并易于工业生产。
【Abstract】 Objective:To study a new synthesis process for preparing salmeterol xinafoate.Methods: Started from salicylaldehyde and 4-phenyl-1-butanol,the target compound was synthesized from salicylaldehyde via several steps involving acetylization,condensation,reduction,and catalytic hydrogenolysis.Results: The total yield was 31.5%.Structures of the target product and key intermediates were verified by IR and 1H-NMR.Conclusion: This optimal synthetic procedure has the advantages of lower cost and higher yield,and is worthy to further industrialized manufacture.
