活血渗湿方对肝纤维化大鼠肝组织中Smad4和Smad7表达的影响
Effects of Huoxue Shenshi decoction on expression of Smad4 and Smad7 in liver of rats with liver fibrosis
【摘要】 目的:探讨活血渗湿方抗肝纤维化的作用机制。方法:应用40%CC l4油剂诱导50只SD大鼠形成肝纤维化模型,随机分成3组,即模型组、渗湿方组和软肝片组,分别灌服0.85%氯化钠溶液、活血渗湿方水煎剂、复方鳖甲软肝片水溶液。另设正常大鼠20只,作为正常组。药物干预12周后,处死大鼠,行肝组织苦味酸-天狼红染色,观察肝纤维化程度;免疫组织化学法检测肝组织Sm ad4和Sm ad7的表达,放射免疫法检测血清透明质酸(HA)水平。结果:与模型组相比,渗湿方组和软肝片组大鼠血清HA水平明显降低(P<0.05)。模型组大鼠肝纤维程度较高,渗湿方组与软肝片组大鼠肝脏纤维化程度较模型组显著改善(P<0.05)。与模型组相比,渗湿方组和软肝片组大鼠肝组织Sm ad4的阳性表达面积明显减少(P<0.05),而Sm ad7的阳性表达面积明显增加(P<0.05)。结论:活血渗湿方具有明显抗肝纤维化作用,其机制可能是调节肝组织Sm ad4和Sm ad7的表达。
【Abstract】 Objective:To explore the mechanism of anti-liver fibrosis effects of Huoxue Shenshi decoction(HSD).Methods:Liver fibrosis was successfully established in 50 rats by subcutaneous injection of carbon tetrachloride(CCl4).Then model rats were divided into three groups randomly,control group,Fufang Biejia Ruangan tablet(FBRT)treatment group and HSD treatment group,and sodium chloride,FBRT and HSD were administered respectively to three groups by gavage everyday.Meanwhile,another 20 rats were arranged as normal group and were given sodium chloride by gavage everyday.After drugs intervention of 12 weeks,all rats were killed.The degree of liver fibrosis was observed through Picric Acid-Sinus Red staining of liver tissue;the expressions of Smad4 and Smad7 in liver tissue were determined with immunohistochemical methods;The content of HA in serum was determined with radio-immunity method.Results:The detection results of HA in serum showed that the content of HA in serum of FBRT and HSD treatment group decreased significantly compared with control group(P<0.05).The classification results of liver fibrosis showed that the degrees of liver fibrosis of rats in FBRT and HSD treatment group decreased significantly compared with control group(P<0.05).The detection results of immunohistochemistry showed that the expressions of Smad4 in liver tissue in FBRT and HSD treatment group decreased obviously compared with control group(P<0.05),and the expressions of Smad7 increased obviously.Conclusion:HSD has obvious anti-liver fibrosis effect and the mechanism of this effect might be regulate expression of Smad4 and Smad7 in liver tissue.
