高分子支架复合骨髓源成骨细胞体外培养的研究
Experimental study on combined culture in vitro of macromolecular scaffold and bone marrow-originated osteoblast
【摘要】 目的观察高分子支架对成骨细胞的增殖和成骨活性的影响。方法将新西兰幼兔骨髓基质细胞经分离、体外培养及诱导获得的成骨细胞接种在盘状LDIG高分子支架上复合培养,同时以多孔聚乙醇酸材料作为对照,通过细胞计数、Ⅰ型胶原、碱性磷酸酶活性和扫描电子显微镜等手段检测成骨细胞增殖数量及成骨活性。结果骨髓源成骨细胞在高分子支架上生长良好,其增殖数量及成骨活性均优于对照组。结论用高分子材料制备的组织工程支架具有理想的多孔网状结构和良好的骨细胞相容性,可以用于构建组织工程骨。
【Abstract】 【Objective】To observe the effect of macromolecular scaffold on the proliferation and ossification activity of bone marrow-originated osteoblast. 【Methods】Osteoblast of young New Zealand rabbit, which were originated from marrow stroma cell via isolation, cultured in vitro and derivated, were inoculated in the macromolecular scaffold prepared with lysine disocyanate-glycerol polymer (LDIG) for combinating culture with porous polyglycolic acid (PGA) scaffold as contrast. Subsequently, the proliferating osteoblast amount and ossification activity were detected by means of cell count,Ⅰ-type collagen and alkaline phosphatase activities (APA) test and scanning electron microscope (SEM). 【Results】Bone marrow-originated osteoblast prosperously proliferated in the macromolecular scaffold and the proliferating osteoblast amount and ossification activity was higher than that in the PGA scaffold. 【Conclusion】The tissue engineering scaffold prepared with macromolecular material is capable of constructing tissue engineering bone because of its ideal porous network structure and favourable osteoblastic compatibility.
