视网膜缺血再灌注损伤骨髓间充质干细胞移植途径的探讨
Approach of injury caused by retinal ischemia-reperfusion in bone marrow-derived mesenchymal stem cells′ transplantation ways
【摘要】 目的探讨骨髓间充质干细胞(MSCs)移植治疗大鼠视网膜缺血再灌注损伤的最佳途径。方法Sprague-Dawley大鼠48只,随机分为正常对照组(CON)、视网膜缺血再灌注损伤(IR)组、视网膜下移植(ST)组与玻璃体腔内移植(VT)组,12只/组。缺血再灌注损伤后6h、24h、72h、7d、14d和28d,动态检测各组ERGb波;缺血再灌注损伤后28d,采用HE与尼氏染色法检测各组RGC密度及内层视网膜厚度的变化。结果IR组缺血再灌注后,ERGb波下降,缺血再灌注后72h,IR组ERGb波降到最低幅值,缺血再灌注后28d,ST组与VT组ERGb波高于IR组(P<0.01);HE染色光镜下观察,IR组视网膜神经节细胞减少,内层视网膜变薄,ST组与VT组节细胞层神经节细胞较多,视网膜内层较厚;IR组视网膜神经节细胞密度较小,内层视网膜厚度较薄,均低于ST组与VT组(P<0.01),ST组视网膜神经节细胞密度与内层视网膜厚度均高于VT组(P<0.05)。结论骨髓MSCs视网膜下移植与玻璃体腔内移植均能显著减轻视网膜缺血再灌注损伤,骨髓MSCs视网膜下移植治疗效果较为显著。
【Abstract】 【Objective】To explore the best way of transplantation of bone marrow mesenchymal stem cells (MSCs) into the retina to treat the ischemic reperfusion (IR) retinal injury. 【Methods】Forty-eight Sprague-Dawley rats were randomly divided into the normal control group (CON, n =12), ischemic reperfusion retinal injury group (IR, n =12), subretinal transplantation group (ST, n =12) and vitreous transplantation group (VT, n =12). B-wave of ERG was dynamically examined in each group at 6, 24, 72 hours, 7, 4, and 28 days after ischemia-reperfusion. 28 days after ischemia-reperfusion, HE and Nissl staining were used to observe the changes of retina and examine the changes of density of RGCs and thickness of inner retina. 【Results】In the IR group, b-wave of ERG decreased after ischemia reperfusion and 72 h after ischemia reperfusion, it decreased to the lowest. 28 d after ischemia reperfusion, b-wave of ERG of both ST group and VT group was higher, as compared with the IR group (P <0.01). The RGC decreased and inner retina was thinner 28 days after IR in the IR group under light microscope, as compared with the ST and VT group (P <0.01). Nissl staining results showed that the density of RGCs decreased and the inner retina was thinner as compared with the ST group and the VT group (P <0.01). And more RGCs and thinner innerretina were observed in the retina of ST group as compared with the VT group (P <0.05). 【Conclusion】Both subretinal transplantation and vitreous transplantation of bone MSCs were capable of attenuating the ischemic reperfusion retinal injury. The subretinal transplantation of bone MSCs was more effective.
