叶酸受体在鼻咽癌组织及细胞中的表达与意义

【摘要】 目的研究人鼻咽癌组织和细胞株中叶酸受体表达情况,探讨其与肿瘤临床和病理特征的关系,为进一步研发叶酸受体靶向抗鼻咽癌药物载体奠定基础。方法分别以免疫组织化学法和免疫细胞荧光法观察20例鼻咽癌组织和体外培养的HNE1、HNE2,CNE1,CNE2,SUNE1,5-8F等6株鼻咽癌细胞株表达叶酸受体的情况,分析鼻咽癌组织叶酸受体表达与临床病理特征的关系。结果鼻咽癌组织叶酸受体阳性表达率为85%(17/20),正常鼻咽黏膜组织中无叶酸受体表达(0/8)。鼻咽癌叶酸受体表达与患者性别、年龄等因素无相关性,在非角化鳞癌和鼻咽未分化癌两种病理类型中无明显差异,但临床分期Ⅲ~Ⅳ期的病例阳性表达高于Ⅱ期以下的病例。HNE1细胞株叶酸受体表达阳性,其余5株细胞为阴性。结论鼻咽癌组织广泛高表达叶酸受体,这为开发针对鼻咽癌的叶酸受体靶向药物奠定了基础。

【Abstract】 【Objective】 In order to investigate the expression of folate receptor in nasopharyngeal carcinoma(NPC) and its correlation with clinical and pathological features.【Methods】Using immunohistochemistry and im-munocytofluorescense detect the expression of folate receptor in 20 NPC specimens and six cell lines such as HNE1,HNE2,CNE1,CNE2,SUNE1,5-8F,the relationship between the expression of FR and clinicopathological features of specimens was analyzed.【Results】The positive expression of folate receptor was detected in 85%(17/20) NPC tissue and negative in normal nasopharyngeal tissue(0/8) which did not correlate with gender,age,pathology type,but correlate with clinical stage(P =0.045).In six cell lines folate receptor positively expression was found only in HNE1.【Conclusions】The results suggested that over express folate receptor is presented in most of NPC tissues,which provided basement to create molecular drug targeting folate receptor on NPC cells.
